新疆哈萨克族ESCC组织LOXL2表达及临床意义

【摘要】:目的哈萨克族食管癌属于新疆地区特高发疾病,其发病率及死亡率均高于全国平均水平。近来越来越多的研究表明,赖氨酰氧化酶样蛋白2(lysyl oxidase-like 2,LOXL2)与肿瘤的侵袭、转移及不良预后有密切关系。本研究探讨LOXL2在新疆哈萨克族食管鳞癌(esophageal squamous cell carcinoma,ESCC)患者中的表达与临床病理特征相关性及与预后的关系。方法检索Oncomine数据库中LOXL2mRNA在ESCC患者中的表达情况,收集2009-03-01-2012-03-31新疆医科大学附属肿瘤医院胸外科80例新疆哈萨克族ESCC患者手术切除标本,免疫组织化学检测LOXL2在80例新疆哈萨克族ESCC患者中蛋白表达,并分析其表达与临床病理特征及总生存时间(overall survival,OS)的关系。结果 Oncomine数据库中共有2个研究涉及ECT2在ESCC组织和正常组织中mRNA表达,2组微阵列数据集中,LOXL2mRNA在ESCC组织中的转录水平均显著高于正常组织,P=3.95×10~(-16),P=6.87×10~(-7)。80例ESCC组织标本及癌旁组织LOXL2蛋白阳性表达率分别为55.0%和28.7%(χ2=4.665,P=0.031),且LOXL2蛋白表达与ESCC的肿瘤分化程度(χ~2=9.126、P=0.010)、淋巴结转移(χ~2=4.393、P=0.036)及TNM分期(χ~2=6.519、P=0.011)呈正相关,与患者性别、年龄、浸润深度及CEA水平无明显相关性,P0.05。LOXL2阳性表达阴性表达的患者5年OS分别为6.7%和35.8%,差异有统计学意义,χ~2=4.612,P=0.032。结论 LOXL2蛋白在新疆哈萨克族ESCC组织中表达明显增高,其蛋白阳性表达与肿瘤分化程度、淋巴结转移及TNM分期密切相关,LOXL2在新疆哈萨克族ESCC的发生发展中起重要作用,可以作为反映新疆哈萨克族食管癌生物学行为及预后的有效指标。
[Abstract]:Objective the incidence and mortality of esophageal cancer in Kazak nationality are higher than the national average. More and more studies have shown that lysyl oxidase like protein 2lysyl oxidase-like 2LOXL2) is associated with tumor invasion. This study was designed to investigate the relationship between the expression of LOXL2 and clinicopathological features and prognosis in Xinjiang Kazak patients with esophageal squamous cell carcinoma. Methods the relationship between LOXL2mRNA and prognosis in Oncomine database was searched. Expression in patients, The specimens of 80 patients with ESCC of Kazak nationality in Xinjiang were collected from thoracic surgery department of Cancer Hospital affiliated to Xinjiang Medical University 2009-03-01-2012-03-31. The protein expression of LOXL2 was detected by immunohistochemistry in 80 cases of ESCC in Xinjiang Kazak nationality. The relationship between the expression of ECT2 and clinicopathological features and total survival time (OS) was analyzed. Results there were two studies on the expression of ECT2 in ESCC and normal tissues. Two groups of microarray data sets of mRNA expression in ESCC tissues were involved in this study. The positive expression rates of LOXL2 protein in ESCC tissues and adjacent tissues were 55.0% and 28.70.The expression of LOXL2 protein was positively correlated with the degree of differentiation of ESCC (蠂 ~ 29.126), lymph node metastasis (蠂 ~ 2 ~ 24.393P ~ (0.036)) and TNM stage (蠂 ~ (26.519) P 0.011), and the expression of LOXL2 protein was significantly higher than that of normal tissues (P ~ (3.95) 脳 10 ~ (-16)), and the positive expression rate of LOXL2 protein was significantly higher than that in normal tissues (蠂 ~ (2.95) 脳 10 ~ (-16)), and the positive rates of LOXL2 protein expression in ESCC tissues and adjacent tissues were 55.0% and 28.7 respectively (蠂 ~ (2) 4.665). The expression of LOXL2 protein was positively correlated with the degree of differentiation of ESCC (蠂 ~ 29.126). There was no significant correlation with sex, age, depth of infiltration and CEA level in the patients with negative expression of P0.05.LOXL2 in 5 years (P < 0.05). There was significant difference in the expression of LOXL2 protein in the ESCC tissues of Kazak nationality in Xinjiang, and the difference was statistically significant. Conclusion the expression of LOXL2 protein in the ESCC tissues of Kazakh nationality in Xinjiang is obviously higher than that in the control group (P < 0.05). The positive expression of Lox2 protein was closely related to tumor differentiation, lymph node metastasis and TNM staging, and played an important role in the occurrence and development of ESCC in Kazak nationality in Xinjiang. It can be used as an effective index to reflect the biological behavior and prognosis of Kazak esophageal cancer in Xinjiang.
