FISH检测C-myc及PSCA在早期前列腺癌诊断中的应用

【摘要】:目的:通过FISH检测C-myc和PSCA在前列腺癌和前列腺增生组织中的表达情况,分析C-myc和PSCA在前列癌组织和前列腺增生组织中的表达情况以及与前列腺癌的临床分期的关系,探讨C-myc和PSCA在早期前列腺癌诊断中的应用。方法:选择2013年12月-2016年10月昌乐县人民医院泌尿外科前列腺患者病理标本55例,其中前列腺癌标本为25例,前列腺增生标本30例。患者术前未经过任何的放化疗。利用FISH方法检测55例前列腺组织中的C-myc和PSCA的表达情况。使用SPSS16.0软件进行Pearson相关性分析,研究二者的表达与前列腺癌的诊断、临床分期之间的关系。结果:C-myc基因的阳性表达率与前列腺细胞学及组织学的病变程度呈现出正相关(P0.001),随着前列腺细胞学及组织学的恶变程度加重而有明显增加(P0.001)。PSCA的表达在前列腺癌组和前列腺增生组中的阳性表达率并没有明显的差异(P0.05),PSCA在前列腺癌和前列腺增生组中的强阳性表达率,二者之间的差异有统计学意义(P0.05)。结论:1、C-myc基因与前列腺病变程度呈现出正相关关系,可作为预测前列腺病变发展的重要指标,可作为早期前列腺癌和高度可疑前列腺癌病变的指标。2、PSCA强阳性表达率在前列腺癌组中明显高于前列腺增生组,并且前列腺癌组中的PSCA表达水平上临床分期有相对较显著的相关性,提示出PSCA对前列腺癌诊断及预后判断有相当的价值。3、结合C-myc基因与PSCA对前列腺癌诊断可增加对于诊察判断的敏锐度。
[Abstract]:Objective: to detect the expression of C-myc and PSCA in prostate cancer and prostatic hyperplasia by FISH, and to analyze the expression of C-myc and PSCA in prostatic carcinoma and prostatic hyperplasia, and the relationship between C-myc and PSCA and the clinical stage of prostate cancer. To explore the application of C-myc and PSCA in the diagnosis of early prostate cancer. Methods: from December 2013 to October 2016, 55 patients with prostate cancer were selected from the urological department of Changle people's Hospital, including 25 patients with prostate cancer. The expression of C-myc and PSCA in 55 cases of prostatic tissue was detected by FISH method. The correlation analysis of Pearson was performed with SPSS16.0 software. To study the expression of both and the diagnosis of prostate cancer, Results there was a positive correlation between the positive expression rate of C myc gene and the pathological changes of prostate cytology and histology (P 0.001). The positive expression rate of C myc gene was significantly increased with the degree of malignancy of prostate cytology and histology. There was no significant difference in the positive expression rate of P0.05 and PSCA between prostate cancer and prostatic hyperplasia, and there was no significant difference in the positive expression rate of P0.05 and PSCA between prostate cancer and benign prostatic hyperplasia. Conclusion there is a positive correlation between the C-myc gene and the degree of prostatic disease, which can be used as an important index to predict the development of prostatic disease. The positive rate of PSCA in prostate cancer group was significantly higher than that in prostatic hyperplasia group. There was a relatively significant correlation between the clinical stage and the expression of PSCA in prostate cancer. It is suggested that PSCA has a significant value in the diagnosis and prognosis of prostate cancer. Combination of C-myc gene and PSCA can increase the sensitivity of diagnosis and judgment of prostate cancer.
