皮细胞从静止状态变成具有迁移能力的顶细胞(tipcell),同时激活间质金属蛋白酶(matrixmetalloproteinase,MMP2或MMP9)降解血管外基质,帮助顶细胞向周围扩增和迁移形成柱状丝状伪足,并逐步形成血管腔(lumen)。最后,周细胞(pericytecell,PC)和血管平滑肌细胞(vascularsmoothmusclecell,SMC)等支持细胞(muralcell)环绕在内皮细胞周围形成完整的新生血管(图2)。
