MiR-205抑制ASPP2表达对宫颈癌的转移及化疗敏感性的影响

【摘要】:为进一步了解MiR-205在宫颈癌中对上皮间质转化(epithelial-mesenchymal transition,EMT),以及细胞增殖和药物敏感性的作用,通过实时荧光定量PCR(quantitative real-time polymerase chain reaction,qRT-PCR)和免疫印迹(Western blotting)对MiR-205的靶蛋白——ASPP2蛋白及EMT相关蛋白标志物的表达进行了分析。同时,研究通过噻唑蓝(MTT)实验和流式细胞技术探究了MiR-205对宫颈癌细胞的增殖和化疗敏感性的影响。结果表明,MiR-205可以通过靶向抑制ASPP2的表达,促进宫颈癌细胞的增殖和对紫杉醇的耐药性。研究发现了MiR-205/ASPP2可能是宫颈癌的治疗和潜在生物标志物。
[Abstract]:In order to further understand the role of MiR-205 in epithelial-mesenchymal transition, cell proliferation and drug sensitivity in cervical cancer,The expression of MiR-205 target protein ASPP2 and EMT related protein markers were analyzed by real-time PCR(quantitative real-time polymerase chain reactionation qRT-PCR and Western blotting.At the same time, the effects of MiR-205 on the proliferation and chemosensitivity of cervical cancer cells were investigated by MTT assay and flow cytometry.The results showed that MiR-205 could promote the proliferation and taxol resistance of cervical cancer cells by targeting the expression of ASPP2.The study found that MiR-205/ASPP2 may be a potential biomarker and treatment for cervical cancer.
