PTEN在口腔白斑及口腔鳞状细胞癌中的表达及其临床意义

【摘要】:目的:研究第10号染色体缺失的磷酸酶及张力蛋白同源基因蛋白(PTEN)在口腔白斑和口腔鳞状细胞癌中的表达及其临床意义,探究其在口腔癌变中可能的机制作用。方法:收集20例口腔鳞状细胞癌组织、20例距癌组织2cm的癌旁正常组织及20例口腔白斑组织,Western blotting测定上述3种组织中PTEN蛋白含量,qRT-PCR测定PTEN mRNA的表达水平,免疫组织化学测定PTEN蛋白在上述组织细胞中的组织学定位与表达情况。结果:1、免疫组化结果显示PTEN蛋白多位于细胞浆中呈黄色、棕色染色。Western blotting、实时荧光定量PCR、免疫组织化学检测发现PTEN蛋白和mRNA在口腔鳞状细胞癌、口腔白斑、正常粘膜组织中的表达逐渐增高(正常粘膜组织口腔白斑口腔鳞状细胞癌),差异具有统计学意义(P0.05),三种试验方法结果较为一致。2、无异常增生白斑PTEN蛋白表达明显高于轻度异常增生或中度异常增生,差异具有统计学意义(P0.05)。3、PTEN蛋白在无淋巴结转移口腔鳞状细胞癌中的表达明显高于有淋巴结转移口腔鳞状细胞癌(P0.05)。4、PTEN蛋白在高分化鳞癌中的表达明显较中低分化鳞癌高(P0.05)。5、不同年龄、不同性别、不同发生部位口腔鳞状细胞癌的PTEN蛋白表达无明显差异(P0.05)。结论:1、PTEN在口腔白斑、口腔鳞状细胞癌和口腔正常组织中存在不同程度的表达,表明PTEN可能是口腔鳞癌发生、发展、侵袭转移过程中的一个重要因素。2、PTEN可能作为口腔癌变早期新的生物标记物,用于口腔鳞癌的诊断、预后判断和癌前病变恶性转化的风险评估,PTEN在临床治疗过程中可能会成为口腔鳞状细胞癌的治疗新靶点。
[Abstract]:Aim: to investigate the expression and clinical significance of PTEN in oral leukoplakia and oral squamous cell carcinoma, and to explore the possible mechanism of PTEN in oral carcinogenesis. Methods: 20 cases of oral squamous cell carcinoma (OSCC) and 20 cases of normal tissues adjacent to 2cm and 20 cases of oral leukoplakia were collected to detect the expression of PTEN protein and PTEN mRNA by qRT-PCR in 20 cases of oral squamous cell carcinoma tissues and 20 cases of oral leukoplakia tissues. The histological localization and expression of PTEN protein were detected by immunohistochemistry. Results the results of immunohistochemistry showed that PTEN protein was mostly yellow in cytoplasm, brown staining, Western blotting, real-time fluorescence quantitative PCR, PTEN protein and mRNA were found in oral squamous cell carcinoma and oral leukoplakia by immunohistochemistry. The expression of PTEN protein in normal mucosa tissues was increased gradually (the difference was statistically significant (P 0.05). The results of the three test methods were consistent. 2. 2. There was no abnormal proliferation of leukoplakia PTEN protein expression. Higher than mild dysplasia or moderate dysplasia, The expression of P0.05PTEN protein was significantly higher in oral squamous cell carcinoma without lymph node metastasis than that in well-differentiated squamous cell carcinoma with lymph node metastasis. The expression of PTEN protein in well-differentiated squamous cell carcinoma was significantly higher than that in well-differentiated squamous cell carcinoma. There was no significant difference in the expression of PTEN protein in oral squamous cell carcinoma (OSCC) with different sex and location (P 0.05). Conclusion the expression of PTEN in oral leukoplakia, oral squamous cell carcinoma and oral normal tissue is different, suggesting that PTEN may be the occurrence and development of oral squamous cell carcinoma. PTEN, an important factor in the process of invasion and metastasis, may be used as a new biomarker for the diagnosis of oral squamous cell carcinoma. Prognostic evaluation and risk assessment of malignant transformation of precancerous lesions PTEN may be a new target for the treatment of oral squamous cell carcinoma (OSCC).
