EFEMP2在子宫内膜癌组织及细胞中的表达及其临床意义

【摘要】：目的探讨包含表皮生长因子的fibulin类细胞外基质2(EFEMP2)在子宫内膜癌组织以及子宫内膜癌细胞中的表达及其临床意义。方法用免疫组织化学法检测200例子宫内膜癌组织以及40例正常子宫内膜组织中EFEM P2的表达;采用免疫细胞化学法、qRT-PCR、Western blotting检测EFEM P2在4种子宫内膜癌细胞(KLE、HEC-1A、HEC-1B、Ishikaw a)中的表达水平;运用生长曲线及软琼脂克隆形成实验检测子宫内膜癌细胞的增殖能力;运用Transwell体外侵袭及转移实验检测子宫内膜癌细胞的侵袭及转移能力。结果 EFEMP2在子宫内膜癌组织、细胞中的表达明显低于正常子宫内膜组织及细胞(P0.001),且与子宫内膜癌晚期、分化程度低、淋巴结转移密切相关(P0.05)。KLE和HEC-1A的增殖、侵袭、转移能力高于HEC-1B和Ishikawa(P0.001)。结论EFEM P2的表达与子宫内膜癌临床分期、组织分化程度、有无淋巴结转移和侵袭生长等恶性表型密切相关,并可能参与抑制子宫内膜癌侵袭、转移过程。
[Abstract]:Objective to investigate the expression and clinical significance of fibulin extracellular matrix 2 (EFEMP2) containing epidermal growth factor (ECM) in endometrial carcinoma and endometrial cancer cells. Methods immunohistochemical staining was used to detect the expression of EFEM P2 in 200 endometrial carcinoma tissues and 40 normal endometrium tissue; immunocytochemistry was used. The expression level of EFEM P2 in 4 endometrial carcinoma cells (KLE, HEC-1A, HEC-1B, Ishikaw a) was detected by method, qRT-PCR and Western blotting. The proliferation ability of endometrial carcinoma cells was detected by the growth curve and the soft agar clone formation test. The invasion and metastasis of endometrial carcinoma cells were detected by the Transwell invasion and metastasis test. Results EFEMP2 was significantly lower than normal endometrium and cells (P0.001) in endometrial carcinoma tissue, and with advanced endometrium, low differentiation, lymph node metastasis closely related (P0.05).KLE and HEC-1A proliferation, invasion, metastasis ability higher than HEC-1B and Ishikawa (P0.001). Conclusion the expression of EFEM P2 and the uterus of Ishikawa (P0.001). The clinical stage of endometrial carcinoma and the degree of tissue differentiation are closely related to the malignant phenotype of lymph node metastasis and invasive growth, and may be involved in inhibiting the invasion and metastasis of endometrial carcinoma.
