microRNAs在胸腺上皮性肿瘤中的差异表达

【摘要】:目的微小RNA(microRNAs)在多种肿瘤发生发展中具有重要作用,但在胸腺上皮性肿瘤中的研究较少。文中旨在研究microRNAs在胸腺上皮性肿瘤(B3型胸腺瘤与胸腺癌)中的表达谱及其意义。方法收集南京军区南京总医院病理科2012年1月至2015年1月间的手术切除B3型胸腺瘤3例(对照组)及胸腺鳞状细胞癌3例(病例组),做基因芯片研究。将病例组与对照组数据相比,计算出各miRNA的差异倍数,筛选出上调与下调的miRNA。通过预测网站预测其靶基因,结合文献检索出与胸腺相关的基因。结果 B3型胸腺瘤与胸腺鳞状细胞癌对比发现,32种差异表达miRNAs在胸腺癌中上调,包括miR-125b-1-3p、miR-3175、miR-4462等;19种差异表达miRNAs在胸腺癌中下调,包括miR-361-5p、miR-130a-3p、miR-3651等。miR-377-5p的靶基因包括AKT1、C9、CD19、CDC42、LSS、MYC;miR-485-5p的靶基因包括ADCYAP1R1、ASPA、CAD、CD63等;miR-183-5p的靶基因包括AKAP12、CD28、FOXP1、MDM4等。结论筛选出B3型胸腺瘤与胸腺癌中的差异表达miRNAs,通过预测软件预测这些miRNAs的靶基因,可以为胸腺上皮性肿瘤的进一步治疗提供了有效的依据。
[Abstract]:Objective RNA (microRNAs) plays an important role in the development of a variety of tumors, but there are few studies in the thymic epithelial tumors. The purpose of this study was to study the expression and significance of microRNAs in the thymic epithelial tumor (B3 type thymoma and thymic carcinoma). Methods the pathology department of the general Hospital of Nanjing, Nanjing military region, from January 2012 to 2015 1 3 cases of B3 thymoma (control group) and 3 cases of thymic squamous cell carcinoma (case group) were performed in month. Compared with the control group, the difference multiple of each miRNA was calculated, and the up regulation and down regulation of miRNA. were screened to predict the target gene through the forecast website and the genes related to the thymus were retrieved in the literature. Results compared with thymic squamous cell carcinoma, 32 different expressions of miRNAs were up-regulated in thymic carcinoma, including miR-125b-1-3p, miR-3175, miR-4462 and so on. 19 differentially expressed miRNAs were down-regulated in thymic carcinoma, including AKT1, C9, miR-130a-3p, and miR-3651, and.MiR-377-5p, including miR-361-5p, miR-130a-3p and miR-3651. The target genes include ADCYAP1R1, ASPA, CAD, CD63 and so on. The target genes of miR-183-5p include AKAP12, CD28, FOXP1, MDM4 and so on. Conclusion the differential expression of B3 type thymoma and thymic carcinoma is miRNAs. The prediction software can predict the target genes of these miRNAs, which can provide an effective basis for the further treatment of thymic epithelial tumors.
