人线粒体转录终止因子3在胃癌组织中的表达及其意义

【摘要】:目的分析hMTERF3 mRNA和蛋白质在胃癌组织及正常组织中的表达,探讨其与胃癌发生、发展的关系。方法分别采用半定量RT-PCR和免疫组化SP法,检测61例胃癌组织及正常组织中hMTERF3 mRNA和蛋白质表达水平,分析hMTERF3表达与胃癌临床分期、病理分级、肌层浸润和淋巴结转移的关系。结果 hMTERF3 mRNA和蛋白质在胃癌组织中的表达水平显著高于正常胃组织。hMTERF3蛋白在正常胃组织阳性率为24.6%(15/61),胃癌组织中的阳性率为86.9%(53/61),差异具有统计学意义。hMTERF3蛋白的表达与胃癌临床分期、病理分级、肌层浸润和淋巴结转移有关。结论 hMTERF3蛋白的表达异常可能是胃癌发生、发展的分子机制之一,可为预测胃癌的发生提供可能的生物学标志物,也可为胃癌基因诊断和基因治疗提供潜在的靶标。
[Abstract]:Objective to analyze the expression of hMTERF3 mRNA and protein in gastric cancer tissues and normal tissues, and to explore the relationship between hMTERF3 mRNA and gastric carcinogenesis and development. Methods Semi-quantitative RT-PCR and immunohistochemical SP method were used to detect the expression of hMTERF3 mRNA and protein in 61 cases of gastric cancer and normal tissues. The relationship between the expression of hMTERF3 and clinical stage, pathological grade, myometrial infiltration and lymph node metastasis of gastric cancer was analyzed. Results the expression of hMTERF3 mRNA and protein in gastric cancer was significantly higher than that in normal gastric tissue (24.615 / 61) and in gastric cancer (86.9% / 53 / 61). The difference was statistically significant in the expression of hMTERF3 protein and the clinical stage of gastric cancer. Pathological grade, myometrial invasion and lymph node metastasis were related. Conclusion the abnormal expression of hMTERF3 protein may be one of the molecular mechanisms of gastric carcinogenesis and development, which may provide a possible biomarker for predicting the occurrence of gastric cancer, as well as a potential target for gene diagnosis and gene therapy of gastric cancer.
