人结直肠癌组织中乙酰胆碱相关受体α9nAChR的表达及临床意义

【摘要】：目的:结直肠癌(colorectal cancer,CRC)是消化系统最常见的恶性肿瘤之一,肿瘤转移与复发是其主要死亡原因。α9nAChR是胆碱能受体(Acetylcholine receptors,AChRs)家族的成员之一,在某些恶性肿瘤中过量表达,并与肿瘤的发生、发展、侵袭、转移过程密切相关。但α9nAchR在结直肠癌中的意义尚不清楚。本研究以采用结直肠癌患者为研究对象,检测结直肠癌组织中α9nAChR的表达情况,并分析其表达与肿瘤临床病理特征的关系。方法:采用免疫组化法(S-P法)检测结直肠癌患者手术切除标本α9nAChR蛋白的表达情况。所取标本均为结直肠癌原发病例,术前均经病理诊断为腺癌,而且患者术前未接受放、化疗等治疗。同期入组病例46例,其中男性25例,女性21例。TNM分期:Ⅰ期7例,Ⅱ期19例,Ⅲ期20例。另取距瘤旁5cm以上的正常结肠组织做对照。标本经4%甲醛溶液固定。结果:1α9nAChR的表达主要定位于肿瘤细胞的细胞质和细胞膜上,一般为棕黄色或者棕褐色,在肿瘤间质的三级淋巴组织中也有表达。α9nAChR在正常结直肠组织中几乎不表达,而在结直肠癌组织中的总表达率为63.04%。2 60岁以下患者标本中α9nAChR的表达阳性率为33.33%,而在60岁以及60岁以上患者标本中α9nAChR的表达阳性率为63.64%,两者具有显著统计学差异(P0.05)。3α9nAChR的表达随着分期的进展而增高,各分期之间阳性表达率无明显统计学差异,其表达与T分期有相关性,并随着淋巴结转移范围增加,α9nAChR的表达的阳性率也随之增加。4在CEA正常的患者中,α9n AChR表达的阳性率为33.33%;在CEA增高(CEA≥5μg/L)的患者,其α9nAChR表达的阳性率为63.64%;两者间存在显著统计学差异(P0.05)。5α9nAChR的表达水平与结直肠癌患者的性别、肿瘤部位、血小板水平均无关。结论1α9nAChR在结直肠癌组织及淋巴组织中表达阳性。2α9nAChR表达与结直肠癌患者的年龄、浸润深度、淋巴结转移以及CEA水平相关,可能参与了肿瘤的进展。
[Abstract]:Objective: colorectal cancer CRC is one of the most common malignant tumors in the digestive system. Tumor metastasis and recurrence are the main causes of death. 伪 9nAChR is a member of the cholinergic receptor acetylcholine receptor AChRs family, which is overexpressed in some malignant tumors. It is closely related to the occurrence, development, invasion and metastasis of tumor. However, the significance of 伪-9nAchR in colorectal cancer is unclear. In this study, the expression of 伪 9nAChR in colorectal cancer was detected and the relationship between the expression of 伪 9nAChR and clinicopathological features of colorectal cancer was analyzed. Methods: the expression of 伪 9nAChR protein in surgical specimens of colorectal cancer patients was detected by immunohistochemical staining (S-P method). All the specimens were primary colorectal cancer, which were diagnosed as adenocarcinoma by pathology before operation, and the patients were not treated with radiotherapy and chemotherapy before operation.
