脑源性神经营养因子-反义长链非编码RNA过表达可抑制肝癌细胞的增殖和侵袭

【摘要】:目的:探讨脑源性神经营养因子-反义长链非编码RNA(brain-derived neurotrophic factor-antisense long non-coding RNA,BDNF-AS)对人肝癌细胞增殖和侵袭能力的影响。方法:将携带BDNF-AS基因的重组慢病毒分别感染肝癌细胞系HepG2和97-H后,采用RT-PCR法检测肝癌细胞中BDNF-AS和BDNF mRNA的表达水平变化。分别采用EdU掺入实验、AnnexinⅤ-FITC/碘化丙啶(propidium iodide,PI)双染和Transwell小室法检测BDNF-AS过表达对HepG2和97-H细胞增殖、凋亡和侵袭能力的影响。结果:感染携带BDNF-AS基因的重组慢病毒后,肝癌HepG2和97-H细胞中BDNF-AS的表达水平均明显升高(P值均0.000 1),而BDNF mRNA的表达水平明显降低(P0.000 1,P=0.001)。BDNF-AS过表达后,HepG2和97-H细胞的凋亡率无明显变化(P值均0.05),但细胞增殖率明显降低(P0.05,P=0.001),侵袭细胞数明显减少(P值均0.000 1)。结论:BDNF-AS过表达可能对肝癌细胞的生长和侵袭具有重要的负调控作用。
[Abstract]:Aim: to investigate the effects of brain-derived neurotrophic factor-antisense long chain noncoding RNA(brain-derived neurotrophic factor-antisense long non-coding RNAs BDNF-ASon on proliferation and invasion of human hepatoma cells.
