原发性肝癌细胞中Rock2调控CEBPD对其增殖功能的作用

【摘要】:目的:探讨Rho相关卷曲螺旋形成蛋白激酶2(Rho associated coiled coil containing proteinkinase 2,Rock2)调控CCAAT增强子结合蛋白Delta(CCAAT/enhancer-binding protein delta,CEBPD)的表达对肝癌细胞增殖功能的影响。方法:1、用荧光定量PCR法检测20例肝癌组织及对应癌旁组织中Rock2和CEBPD的m RNA表达,同时分析它们之间的关联;2、用Western blot法检测20例肝癌组织及对应癌旁组织中Rock2和CEBPD的蛋白表达,同时分析它们之间的关联;3、用荧光定量PCR法观察降低Rock2细胞中CEBPD m RNA表达变化;4、用Western blot法观察降低Rock2细胞中CEBPD蛋白表达变化;5、采用EDU实验和CCK8实验观察无意干扰对照组、下调表达Rock2组HCCLM3、SMMC7721肝癌细胞增殖能力的变化。结果:1、Rock2在肝癌组织表达明显高于癌旁组织,CEBPD在肝癌组织表达明显低于癌旁组织,且两者的表达呈负相关性。2、沉默肝癌细胞中Rock2的表达后发现CEBPD的m RNA和蛋白表达水平上调;3、Rock2通过调节CEBPD表达从而影响肝癌细胞的增殖功能。结论:在原发性肝癌细胞中沉默Rock2可引起CEBPD表达上调,进而抑制肝癌细胞的增殖功能。
[Abstract]:Aim: to investigate the effect of Rho associated crimp forming protein kinase (2(Rho associated coiled coil containing proteinkinase 2) on the proliferation of hepatocellular carcinoma (HCC) cells by regulating the expression of CCAAT enhancer binding protein Delta(CCAAT/enhancer-binding protein delta-CEBPD. Methods the m RNA expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were detected by fluorescence quantitative PCR, and the expression of Rock2 and CEBPD in 20 cases of HCC and its adjacent tissues were analyzed by Western blot method. At the same time, the association between them was analyzed. The changes of CEBPD m RNA expression in Rock2 cells were observed by fluorescence quantitative PCR method. The changes of CEBPD protein expression in Rock2 cells were observed by Western blot assay. The unintentional interference control group was observed by EDU experiment and CCK8 experiment. The proliferation ability of HCCLM3 and SMMC7721 hepatoma cells was down-regulated in Rock2 group. Results the expression of Rock2 in HCC tissue was significantly higher than that in paracarcinomatous tissue (CEBPD), which was significantly lower than that in paracancerous tissue. After silencing the expression of Rock2 in hepatoma cells, it was found that the expression of m RNA and protein of CEBPD was up-regulated by regulating the expression of CEBPD, thus affecting the proliferation of HCC cells. Conclusion: silencing Rock2 can induce up-regulation of CEBPD expression and inhibit the proliferation of HCC cells.
