Arpin在结直肠癌组织中的表达及其临床意义

【摘要】:目的:检测Arpin在结直肠癌及正常结直肠组织中的表达情况,并探讨其与结直肠癌临床病理特征的关系及其异常表达的临床意义。方法:收集2015年3月—2015年12月于青岛市市立医院结直肠外科行结直肠癌根治术,并经术后病理诊断证实为结直肠癌的所有患者,根据入组标准,最终选取80例作为研究对象。根据患者临床病理特征的不同,将80例结直肠癌患者分为不同组。用于实验的结直肠组织取自手术切除标本。另外22例石蜡包埋的正常结直肠组织块(距离癌组织边缘超过5cm)作为对照组。采用免疫组织化学法(IHC)检测Arpin蛋白在结直肠癌组织及正常结直肠组织中的表达,同时运用实时荧光定量PCR(q RT-PCR)分别对80对结直肠癌组织及与其配对的癌旁组织中的Arpin m RNA进行检测。结果:Arpin蛋白在结直肠癌组织中的表达水平明显低于正常结直肠组织(32.50%vs86.36%,?2=20.306,P0.001)。对80例经过免疫组化分析的结直肠癌样本进行卡方分析表明,在有无淋巴结转移(81.82%vs 50.00%,?2=9.138,P=0.003)及远处转移(89.47%vs 60.66%,?2=5.484,P=0.019)的肿瘤组织中Arpin蛋白的表达水平不同,差别具有统计学意义。Arpin蛋白在不同TNM分期的结直肠癌患者的癌组织中表达水平也不同,差别具有统计学意义(?2=26.901,P0.001)。实时荧光定量PCR结果显示,结直肠癌组织中Arpin m RNA的表达水平明显低于与其配对的癌旁组织(P0.001)。结论:结直肠癌组织中Arpin的低表达水平与有无淋巴结转移、远处转移及TNM分期有关,Arpin表达水平的降低可能导致肿瘤侵袭和转移能力的获取。
[Abstract]:Aim: to detect the expression of Arpin in colorectal cancer and normal colorectal tissues, and to explore the relationship between Arpin and clinicopathological features of colorectal cancer and its clinical significance. Methods: from March 2015 to December 2015, all the patients with colorectal cancer proved to be colorectal cancer by pathological diagnosis were collected from Qingdao City Hospital. According to the criteria of admission, 80 patients were selected as the study objects. According to the clinicopathological characteristics of the patients, 80 patients with colorectal cancer were divided into different groups. The colorectal tissue used in the experiment was obtained from surgical specimens. Another 22 cases of paraffin embedded normal colorectal tissue mass (more than 5 cm from the edge of cancer tissue) served as control group. Immunohistochemical method was used to detect the expression of Arpin protein in colorectal cancer tissues and normal colorectal tissues. At the same time, the Arpin mRNA in 80 pairs of colorectal cancer tissues and their matched adjacent tissues were detected by real-time fluorescence quantitative PCRQ RT-PCR. Results the expression of the protein in colorectal carcinoma was significantly lower than that in normal colorectal tissue (n = 32.50). The chi-square analysis of 80 colorectal cancer samples with immunohistochemical analysis showed that the expression of Arpin protein was different in the tumor tissues with or without lymph node metastasis (81.82 vs 50.0029.138P0. 003) and distant metastasis (89.47 vs 60.66484 P0. 019). The expression level of Arpin protein in colorectal cancer tissues with different TNM stages was also different, and the difference was statistically significant (P 0.001). The results of real-time fluorescence quantitative PCR showed that the expression level of Arpin mRNA in colorectal cancer tissues was significantly lower than that in adjacent tissues (P0.001). Conclusion: the low expression level of Arpin is associated with lymph node metastasis, distant metastasis and decreased expression of Arpin in colorectal carcinoma, which may lead to the acquisition of tumor invasion and metastasis.
