【摘要】:目的:研究miRNA-195对U251细胞株CCND1表达的影响,试探miRNA-195与U251周期的关系,从而为临床上总结胶质瘤治疗要领提供一定的基础。方法:通过脂质体Lipofectamin 2000将MiRNA-195mimics、MiRNA-195inhibitor转染至胶质瘤细胞U251,Real-time PCR检测转染前后MiRNA-195的含量以确保转染效率;再通过Real-time PCR检测转染前后CCND1mRNA的表达;Western Blotting检测胶质瘤细胞U251中CCND1的表达水平;应用SPSS13.0统计软件处理数据,定量资料采用(?)±s描述,多组间数据比较采用单因素方差分析,组间两两比较采用LSD-t检验,P0.05为差异有统计学意义。结果:1、转染MiRNA-195mimics的U251胶质瘤细胞中的CCND1mRNA的表达高于转染MiRNA-195inhibitor的U251胶质瘤细胞;2、转染MiRNA-195mimics的U251胶质瘤细胞中的CCND1蛋白的表达高于转染MiRNA-195inhibitor的U251胶质瘤细胞。结论:1、MiRNA-195对胶质瘤U251细胞株CCND1表达有影响;2、过表达MiRNA-195下调胶质瘤U251细胞CCND1的表达。
[Abstract]:Objective: To study the effect of miRNA-195 on the expression of CCND1 in U251 cell line and explore the relationship between miRNA-195 and U251 cycle, so as to provide a certain basis for clinical summary of the treatment of glioma. Method: MiRNA-195mimics, MiRNA-195inhibitor is transferred to U251 in glioma cells through liposome Lipofectamin 2000, and Real-time PCR is used to detect the transfection before transfection. The transfection efficiency was ensured by the content of post MiRNA-195, and the expression of CCND1mRNA was detected before and after transfection by Real-time PCR; Western Blotting was used to detect the expression of CCND1 in glioma cell U251; the data were processed with SPSS13.0 statistical software, and the quantitative data were described with (?) + s, and the multiple groups of data were compared by single factor analysis of variance, 22 between groups. Compared with LSD-t test, the difference was statistically significant. Results: 1, the expression of CCND1mRNA in the U251 glioma cells transfected with MiRNA-195mimics was higher than that of U251 glioma cells transfected with MiRNA-195inhibitor; 2, the expression of CCND1 egg white in the U251 glioma cells transfected with MiRNA-195mimics was higher than that of MiRNA-195inhibitor U251 gum. Tumor cells. Conclusion: 1, MiRNA-195 has an effect on the expression of CCND1 in glioma cell line U251; 2, overexpression of MiRNA-195 down-regulation of CCND1 expression in glioma U251 cells.
