MicroRNA在不同分化程度非小细胞肺癌中差异表达的研究

【摘要】:目的:通过芯片技术筛查不同分化程度非小细胞肺癌中Micro RNA(mi RNA)的差异表达。方法:本研究通过收集宁夏医科大学总医院普胸外科手术切除的肺良性病变远端正常肺组织、低分化非小细胞肺癌和高分化非小细胞肺癌组织各3例,通过提取总RNA后,使用mi RNA芯片和m RNA芯片联合筛查,经过筛选,找到表达差异的mi RNA。结果:经过筛查,找到差异表达的mi RNA共27个,其中相对于正常肺组织,低分化非小细胞肺癌与高分化非小细胞肺癌共同表达上调的mi RNA有9个:hsa-mi R-3651,hsa-mi R-210-3p,hsa-mi R-449c-5p,hsa-mi R-375,hsa-mi R-494-3p,hsa-mi R-501-5p,hsa-mi R-664b-3p,hsa-mi R-19b-3p,hsa-mi R-200a-3p,这其中共同表达上调且分化程度越低表达上调越明显的是:hsa-mi R-3651,hsa-mi R-210-3p。相对于正常肺组织,低分化非小细胞肺癌与高分化非小细胞肺癌中表达共同下调的有14个:hsa-mi R-34c-3p,hsa-mi R-30a-3p,hsa-let-7f-5p,hsa-mi R-30c-2-3p,hsa-mi R-3136-5p,hsa-mi R-34b-3p,hsa-mi R-3911,hsa-mi R-486-3p,hsa-mi R-486-5p,hsa-mi R-6127,hsa-mi R-642a-3p,hsa-mi R-760,hsa-mi R-126-3p,hsa-mi R-1247-3p。这其中共同下调且分化程度越低下调越明显的是:hsa-mi R-34c-3p,hsa-mi R-30a-3p。我们还发现了在高分化非小细胞肺癌组织内下调,低分化非小细胞肺癌组织内上调的mi RNA有:hsa-mi R-28-5p,hsa-mi R-29b-1-5p,hsa-mi R-31-5p。高分化非小细胞肺癌组织内上调,低分化非小细胞肺癌组织内下调的mi RNA有:hsa-mi R-449c-5p。结论:根据我们筛查到的这些mi RNA,其中hsa-mi R-3651,hsa-mi R-210-3p,hsa-mi R-34c-3p,hsa-mi R-30a-3p很有可能与非小细胞肺癌的分化程度有关,而共同上调与共同下调的mi RNA很有可能是与非小细胞有关的促癌或者抑癌因子。而分化程度不同,呈现反向表达的hsa-mi R-28-5p,hsa-mi R-29b-1-5p,hsa-mi R-31-5p,hsa-mi R-449c-5p有可能是分化程度不同的非小细胞肺癌特有的差异mi RNA。
[Abstract]:Objective: to screen the differential expression of Micro RNA (mi RNA) in non-small cell lung cancer (NSCLC) with different differentiation by microarray technique. Methods: in this study, the distal normal lung tissues of benign lung lesions, three cases of poorly differentiated non-small cell lung cancer (NSCLC) and three cases of well-differentiated NSCLC (highly differentiated NSCLC) were collected from thoracic surgery of Ningxia Medical University General Hospital, and the total RNA was extracted. The mi RNA chip and m RNA chip are used to screen and find the mi RNA. with different expression. Results: after screening, 27 differentially expressed mi RNA were found, compared with normal lung tissue.
