FAT10、NF-κBp50在乳腺浸润性导管癌中的表达及其临床意义

【摘要】:[目的]探讨双泛素(human leukocyte antigen F-associated transcript 10,Diubiquitin,FAT10)和核因子 κ Bp50 (nuclear factor κBp50,NF-κBp50)在乳腺浸润性导管癌中的表达情况及其临床病理意义。[方法]本研究选取云南省肿瘤医院2014-2015年间手术切除或穿刺的石蜡标本,其中包括112例乳腺浸润性导管癌组织和53例癌旁组织,应用MaxvisionTM免疫组织化学的方法,检测乳腺浸润性导管癌癌组织和癌旁组织中FAT10与NF- κ B p50的蛋白表达情况;同时收集2016-2017年间新鲜的乳腺浸润性导管癌组织和癌旁组织共28对,应用实时荧光定量PCR的方法,检测乳腺浸润性导管癌癌组织和癌旁组织中FAT10与NF- κ B p50的mRNA水平;并分别从蛋白水平和mRNA水平分析二者与乳腺浸润性导管癌患者临床病理因素之间的关系,以及FAT10与NF- κ B p50二者之间表达的相关性。[结果]FAT10和NF- κ Bp50蛋白在乳腺浸润性导管癌中的阳性表达率分别为77.7%和81.3%,明显高于癌旁组织(39.6%和45.3%),差异有显著统计学意义(P0.001); FAT10和NF-κBp50mRNA在乳腺浸润性导管癌中的相对表达量中位数分别为6.21和4.08,明显高于癌旁组织1,差异有统计学意义(P0.05)。FAT10、NF- κ Bp50蛋白在有淋巴结转移和远处转移的癌组织中高表达,并随组织学分级、TNM分期的升高而表达逐渐升高,差异有统计学意义(P0.05),但与患者年龄、肿瘤大小及分子分型差异无统计学意义(P0.05)。FAT10和NF-κ Bp50的mRNA水平与患者淋巴结转移、远处转移、组织学分级、TNM分期、患者年龄、肿瘤大小及分子分型间差异均无统计学意义(P0.05)。此外,FAT10与NF- κ Bp50两者之间蛋白和mRNA的表达均呈正相关(r=0.621,P0.001 和 r=0.475,P=0.011)。[结论]1、FAT10、NF-κBp50在乳腺浸润性导管癌中的表达高于癌旁组织。2、FAT10、NF-κBp50蛋白在有淋巴结转移和远处转移的癌组织中高表达,并随组织学分级、TNM分期的升高而表达升高,但与患者年龄、肿瘤大小、分子分型差异无统计学意义,提示二者与乳腺癌的恶性程度密切相关,可能参与促进了乳腺癌的进展。3、FAT10、NF-κ Bp50 mRNA水平与临床病理因素之间的关系未显示出统计学意义,可能与表观调控有关,具体原因有待进一步的研究。4、FAT10、NF-xBp50二者在乳腺浸润性导管癌中的表达呈正相关,提示二者可能在乳腺癌的发生发展中协同发挥作用。
[Abstract]:[objective] to investigate the expression and clinicopathological significance of diubiquitin (human leukocyte antigen F-associated transcript 10 diubiquitin (human leukocyte antigen F-associated transcript 10) and nuclear factor- 魏 Bp50 (nuclear factor 魏 Bp50,NF- 魏 Bp50 in breast invasive ductal carcinoma. [methods] A total of 112 breast invasive ductal carcinoma tissues and 53 paracancerous tissues were collected from Yunnan Cancer Hospital from 2014 to 2015. MaxvisionTM immunohistochemical method was used. To detect the expression of FAT10 and NF- 魏 B p50 protein in breast invasive ductal carcinoma tissues and adjacent tissues, 28 pairs of fresh breast invasive ductal carcinoma tissues and adjacent tissues were collected from 2016 to 2017, and real-time fluorescence quantitative PCR was used to detect the expression of FAT10 and NF- 魏 B p50 in breast invasive ductal carcinoma tissues and adjacent tissues. To detect the mRNA levels of FAT10 and NF- 魏 B p50 in breast invasive ductal carcinoma tissues and adjacent tissues, and to analyze the relationship between FAT10 and NF- 魏 B p50 and clinicopathological factors in breast invasive ductal carcinoma from protein level and mRNA level, respectively. And the correlation between the expression of FAT10 and NF- 魏 B p50. [results] the positive expression rates of FAT10 and NF- 魏 Bp50 in breast invasive ductal carcinoma were 77.7% and 81.3%, respectively, which were significantly higher than those in paracancerous tissues (39.6% and 45.3%), the difference was statistically significant (P0.001), and the relative expression of FAT10 and NF- 魏 Bp50mRNA in invasive ductal carcinoma of breast was significantly higher than that of adjacent tissues (39.6% and 45.3%). The median expression levels of FAT10,NF- 魏 Bp50 were 6.21 and 4.08, respectively, which were significantly higher than those in adjacent tissues (P0.05). The expression of FAT10,NF- 魏 Bp50 protein was highly expressed in cancer tissues with lymph node metastasis and distant metastasis. With the increase of histological grade and TNM stage, the expression of mRNA increased gradually, the difference was statistically significant (P0.05), but there was no significant difference with age, tumor size and molecular typing (P0.05). The mRNA levels of FAT10 and NF- 魏 Bp50 were correlated with lymph node metastasis and distant metastasis. There was no significant difference in histological grade, TNM stage, age, tumor size and molecular typing between patients (P0.05). In addition, there was a positive correlation between the expression of protein and mRNA between FAT10 and NF- 魏 Bp50 (r = 0.621, P 0.001, r = 0.475, P = 0.011). [conclusion] 1 the expression of NF- 魏 Bp50 in breast invasive ductal carcinoma is higher than that in adjacent breast tissue. 2FAT10NF-kappa Bp50 protein is highly expressed in cancer with lymph node metastasis and distant metastasis. The expression of NF- 魏 Bp50 is increased with the increase of histological grade and TNM stage, but it is associated with the age of the patients. The difference of tumor size and molecular type was not statistically significant, suggesting that they were closely related to the degree of malignancy of breast cancer, and might be involved in promoting the progression of breast cancer. 3 the relationship between the level of NF- 魏 Bp50 mRNA in FAT10 and clinicopathological factors was not statistically significant. The expression of NF-xBp50 and NF-xBp50 in invasive ductal carcinoma of breast is positively correlated, suggesting that they may play a synergistic role in the occurrence and development of breast cancer.
