射频加热增强前列腺癌局部化疗疗效的实验研究
[Abstract]:Objective to establish a new interventional therapy for enhancing the efficacy of local chemotherapy for prostate cancer with radiofrequency heating (radiofrequency heat, RFH). Materials and methods this experiment is divided into two parts: in vivo experiment and in vitro experiment. In vitro experiment part: human prostate cancer cell line cultured in vitro was divided into 4 groups: (1) control group; (2) radiofrequency heating group only; (3) only chemotherapy group (docetaxel); (4) combined radiofrequency heating and chemotherapy group. After the cells in each group were treated with different methods, the proliferation level of tumor cells in the radiofrequency heating-chemotherapy combined therapy group was compared with that in the other three groups by MTS colorimetry. In vivo part: the nude mice model of human prostate cancer transplanted subcutaneously was firstly established and divided into 4 groups (n = 6): (1) control group, (2) radiofrequency heating group only, (3) chemotherapy group (docetaxel only), 3) chemotherapy group (docetaxel group), 3) chemotherapy group (docetaxel group). (4) radiofrequency heating combined with chemotherapy group. Before and 1 and 2 weeks after treatment, the tumor volume of the transplanted tumor model was monitored by magnetic resonance imaging (MRI). Finally, the apoptosis level of the tumor tissue was confirmed by pathological analysis. Results in the part of in vitro experiment: the proliferation level of human prostate cancer cells in the radiofrequency heating-chemotherapy group was significantly lower than that in the other three groups (P0.05). In vivo experiment part: (1) after radiofrequency heating-chemotherapy combined treatment, the transplanted tumor volume of mice was significantly smaller than that of the other three groups (P0.05); The apoptotic level of transplanted tumor in the combined treatment group was significantly higher than that in the other groups (P0.05). The TUNEL apoptosis in the tumor tissue was significantly higher than that in the other groups. Conclusion radiofrequency heating can enhance the efficacy of local chemotherapy for prostate cancer in vitro and in vivo. The combination of RFH and local chemotherapy has laid a foundation for the removal of tumor tissue in the tumor ablation boundary region. The new interventional therapy may provide a new therapy for prostate cancer patients.
