miR-21在肝癌中的表达及其与PTEN的关系

【摘要】：目的:探讨miR-21在肝癌中的表达及其与PTEN的关系。方法:用RT-PCR检测119例肝癌组织与癌旁组织中miR-21与PTEN的表达情况;双报告基因实验检测肝癌细胞中miR-21对PTEN转录水平的影响;分析miR-21表达与肝癌患者临床病理因素的关系;在肝癌细胞中分别过表达及抑制miR-21的表达之后,检测细胞增殖与PTEN及其下游通路蛋白的表达。结果:肝癌组织中miR-21表达明显高于癌旁组织,而PTEN则相反(均P0.05);miR-21与PTEN的表达具有一定相关性(r2=0.6767,P0.05);miR-21可并抑制PTEN转录活性;miR-21的表达与肝癌患者AFP水平及TNM分期有关(均P0.05);肝癌细胞过表达miR-21后,肝癌细胞增殖明显增强,PTEN表达明显调,并影响其下游蛋白的表达,而抑制miR-21则相反(均P0.05)。结论:miR-21在肝癌中表达升高,且miR-21可以通过下调PTEN表达进一步促进肝癌细胞的增殖。
[Abstract]:Objective: to investigate the expression of miR-21 in hepatocellular carcinoma (HCC) and its relationship with PTEN. Methods: the expression of miR-21 and PTEN in 119 HCC tissues and paracancerous tissues was detected by RT-PCR, the effect of miR-21 on PTEN transcription in HCC cells was detected by double reporter gene assay, the relationship between miR-21 expression and clinicopathological factors was analyzed, and the expression of PTEN and its downstream pathway proteins were detected after overexpression and inhibition of miR-21 expression in HCC cells. Results: the expression of miR-21 in HCC tissues was significantly higher than that in paracancerous tissues, while the expression of PTEN was opposite (all P 0.05).
