B7-H4对肝癌细胞凋亡和自噬的影响及机制研究

目的:B7-H4和自噬可以通过PI3K信号通路调节或诱导。然而,在肝细胞癌中B7-H4与自噬的关系仍未清楚。本课题的目的是研究肝细胞癌中B7-H4是否通过PI3K信号通路调节自噬。方法:siRNA敲低Huh7和Hep3B细胞B7-H4后,采用CCK-8法检测细胞增殖,流式细胞术检测细胞凋亡,western blot检测cleaved-caspase3、cleaved-PARP、BAX、BCL-2、LC3、P62、P-AKT、P-PI3K、P-m TOR和总AKT、PI3K、m TOR蛋白水平,MDC法检测自噬体。结果:敲低Huh7和Hep3B细胞B7-H4后,siRNA组与NC组相比,细胞增殖减少,细胞凋亡率增加;凋亡相关指标cleaved-caspase3、cleaved-PARP、BAX蛋白增加,P62蛋白降低;自噬相关指标LC3Ⅱ增加,Bcl2蛋白降低;MDC法可见siRNA组更多自噬体;AKT、PI3K、m TOR的磷酸化被抑制;siRNA组和NC组分别使用CQ抑制LC3降解后发现LC3Ⅱ仍然增加,使用3MA抑制PI3K后,发现细胞凋亡和自噬减弱。结论:B7-H4通过PI3K信...
