长链非编码RNA H19在食管鳞癌中的表达水平及其临床预后相关性的研究

食管癌是最致命的恶性肿瘤之一。它是导致肿瘤死亡的第六大原因,也是世界上第八大最常见的肿瘤。主要是因为它极难早期诊断,侵袭能力强,因此预后差以及存活率低。其五年存活率约为15%-25%。更糟糕的是,随着世界范围内人口老龄化的增长,食管癌的发病率也正在逐年上升。中国是食管癌发病率最高的地区之一,尤其以中国河南省安阳市最高。长链非编码RNA(long non-coding RNA,lncRNA)为长度大于200个核苷酸,不具有编码蛋白功能的RNA分子。报道表明,它可以通过与DNA、染色质、蛋白质、RNA等大分子相互作用促进或抑制肿瘤的发生、侵袭和转移,其复杂的作用机制是目前研究的重点和难点。据报道,LncRNAH19在成人组织中表达量低或不表达,但在多种肿瘤中高表达,如大肠癌、乳腺癌、膀胱癌等,且与肿瘤的发生发展密切相关。因此LncH19有可能具有肿瘤早期诊断和预后判断的标志物的临床价值。本课题大规模地使用ISH方法检测食管鳞癌中H19的表达水平,旨在探讨其在食管癌中的临床病理作用。其次运用生物信息学分析方法预测LncRNA H19的靶向miRNA,探讨H19在食管鳞癌中可能的作用机制。方法...
