探索长链非编码RNAs的异常表达及肿瘤病理学中的其他分子改变

背景:癌症是重大全球公共卫生问题,预计到2040年癌症患者数量将增加60%以上。世界卫生组织数据显示,2018年全球新增癌症病例1810万例,预计到2040年每年新增病例数将突破2940万例。由于对癌症的遗传和分子基础了解不足阻碍了治疗方案的开发,以及缺乏可靠的癌症诊断和预后生物标志物,导致癌症患者的死亡率居高不下。为了更好地抗击癌症,需要对导致癌变和疾病发展的过程,包括引起肿瘤的分子和遗传改变等进行更详细、深入的探究。因此,本论文的整体目标是发现新的在癌症中失调的分子,检查其在癌症预后中的作用,并揭示其发挥作用机制,从而确定其作为肿瘤生物标志物或治疗靶标的可能性。具体来说,本研究分析了六种长链非编码RNA(PTPRG-AS1,FOXP4-AS1,BLACAT2,ZXF2,UCC和SAP30L-AS1)在胃癌中的表达水平及其临床意义。接下来,研究了 8种不同的CBX家族成员在胃癌中的表达水平和预后判断价值。然后,评估了 VCAN在多种癌症中的表达水平、分子机制及预后判断中的作用。最后,阐明了核糖核苷酸还原酶调节亚基M2(RRM2)在30种不同人类癌症致癌过程中的作用。方法:通过RT-q...
