清除Treg细胞提高SA-mGM-CSF前列腺癌细胞疫苗的抗肿瘤效果

研究背景前列腺癌是欧美国家男性最常见的恶性肿瘤之一,早期局限性前列腺癌一般采用手术或局部放疗,而晚期多采用内分泌治疗,但随着时间的延长,很多患者发展为去势抵抗性前列腺癌。免疫治疗(疫苗或细胞因子)是治疗肿瘤的有效途径之一。本实验将链亲和素标记的mGM-CSF(SA-mGM-CSF)锚定在生物素化的肿瘤细胞表面,成功制备一种新型肿瘤细胞疫苗。研究表明该疫苗具有较好的疗效,但疫苗治疗后肿瘤微环境中的Treg细胞也明显增加。Treg细胞是一群具有免疫抑制功能的T细胞亚群,参与肿瘤免疫逃逸。因此,抑制或清除Treg细胞必将有效提高抗肿瘤免疫反应。近期研究发现,ICOS+Treg是发挥免疫抑制作用的主要Treg亚群。我们前期的研究发现,体外活化ICOS信号可显著促进Treg细胞的增殖和免疫抑制功能。基于以上研究结果,我们提出设想:采用ICOS封闭性抗体靶向清除Treg细胞,从而提高疫苗的疗效。去甲斑蝥素(NCTD)具有很强的抗肿瘤能力,能抑制多种肿瘤细胞的增殖并诱导其凋亡。NCTD具有区别于普通化疗药物的最显著特点,即NCTD具有升白作用,可减少化疗后的骨髓抑制。近期有报道称,NCTD还具有免疫...
