咖啡因拮抗针刺抗炎症痛效应及下丘脑疼痛信号分子的表达

【摘要】:目的:研究咖啡因对炎症痛针刺镇痛效应及下丘脑疼痛相关分子表达的影响。方法:以C57BL/6J小鼠为研究对象,筛选基础热痛阈值在10 s~20 s的小鼠,随机分为空白组、模型组、模型+电针组(简称模针组)、模型+电针+咖啡因组(简称模咖针组);使用完全弗氏佐剂足底注射制备炎症痛模型;模咖针组摄入70 mg/Kg/day咖啡因,空白组、模型组、模针组摄入饮用水;模针组、模咖针组进行电针干预,2/100Hz疏密波,0.2mA,15分钟,1次/日,连续干预11天;采用辐射热测痛法测定各组痛阈,治疗结束后处死动物取下丘脑,用PCR Array高通量检测技术检测下丘脑疼痛相关分子的表达。结果:(1)咖啡因对炎症痛针刺镇痛效应的影响结果:组间比较:造模后3天,与空白组比较,模型组、模针组以及模咖针组小鼠痛阈明显下降,差异有统计学意义(P0.000),而模型组、模针组以及模咖针组三组间差异无统计学意义(P0.05),具有可比性。在造模后7天,模针组与模型组痛阈比较差异有统计学意义(P0.000);模咖针组与模型组痛阈比较差异无统计学意义(P0.05);模咖针组与模针组比较差异有统计学意义(P0.000)。在造模后13与14天,模针组与模型组痛阈比较差异有统计学意义(PO.000),模咖针组与模型组痛阈比较差异无统计学意义(P0.05),模咖针组与模针组比较差异有统计学意义(P0.001)。组内比较:模型组在造模后7天的痛阂与造模后3天比较有所改善(P0.001);在造模13与14天的痛阈与造模后3天比较差异有统计学意义(P0.000);在造模13与14天的痛阈与造模后7天比较差异有统计学意义(PO.000);而第13和14天的痛阈差异无统计学意义(P0.05)。模针组在造模后7天的痛阈与造模后3天比较差异有统计学意义(P0.000);模针组在造模13与14天的痛阈与造模后7天比较异有统计学意义(P0.000);模针组在造模13与14天的痛阈比较差异无统计学意义(P0.05)。模咖针组在造模后7天的痛阈与造模后3天比较差异有统计学意义(P0.05);模咖针组在造模13与14天的痛阈与造模后7天比较差异有统计学意义(P0.001);模咖针组在第13天与第14天痛阈比较差异无统计学意义(P0.05)。(2)下丘脑内疼痛相关分子mRNA的表达:与空白相比较,模型组有18个基因明显,其中表达上调有13个,下调有5个;与模型组比较,模针组基因有7个基因变化明显,其中表达下降有1个,表达上调有6个;与模型组比较,模咖针组有25基因变化明显:表达上调有24个,表达下调有1个;与模针组比较,模咖针组有25基因变化明显,其中表达上调有24个,表达下调有1个。其中,与咖啡因影响电针镇痛相关的基因是Ngf。与空白组相比,模型组中基因Ngf的表达上调,上调倍数为1.9053;与模型组相比,模针组中Ngf基因的表达下调,下调倍数为0.4475;与模型组相比,模咖针组中基因Ngf变化不明显,其值为0.7253。结论:(1)电针对炎症痛有确切的镇痛效应;(2)电针期间摄入咖啡因(70 mg/Kg/day),可抑制电针的镇痛效应;(3)咖啡因摄入减弱电针镇痛效应可能与下丘脑Ngf基因的表达有关。
[Abstract]:Objective: To study the expression effect of caffeine on inflammatory pain and the analgesic effect of acupuncture and pain related hypothalamic molecules.
