【摘要】:目的:探讨“肺肠合治法”治疗干燥综合征的中医理论基础;探讨肺肠合治中药麦冬地芍汤治疗干燥综合征调节Th1/Th2、Th17/Treg免疫失衡及AQP5表达异常是否与其上调神经肽VIP的表达相关。方法:33只雌性NOD小鼠随机分为3组:模型组、羟氯喹组、中药组,8只ICR (Institute of Cancer Research)小鼠为空白组。羟氯喹组、中药组分别灌服羟氯喹溶液及麦冬地芍汤,空白及模型组灌服生理盐水,18周龄时采集颌下腺及血清。检测小鼠10、13、16周龄时的唾液流率,Elisa法检测血清及一侧颌下腺中VIP及细胞因子IFN-γ、IL-10、IL-17A的含量,另一侧颌下腺作HE染色,免疫组化法测AQP5的表达。结果:1.与空白组相比,模型组小鼠唾液流率明显减少,与模型组相比,中药组小鼠在16周龄时唾液流率明显改善(P0.01)。2.中药组小鼠血清及颌下腺中VIP浓度较模型组明显升高(P0.05),IFN-γ、IL-17A水平较模型组降低(P0.01),Thl/Th2、Th17/Treg比值较模型组明显降低(P0.01)。3.颌下腺病理显示,NOD小鼠较正常小鼠颌下腺可见明显淋巴细胞浸润,而经麦冬地芍汤治疗,颌下腺病理评分明显改善(P0.05)。免疫组化显示模型组小鼠AQP5在颌下腺的表达明显减低,羟氯喹及中药组AQP5的表达较模型组上调(P0.05)。结论:麦冬地芍汤对干燥综合征动物模型NOD小鼠有显著疗效,改善了小鼠唾液流率,调节了Thl/Th2、Th17/Treg免疫平衡,改善颌下腺淋巴细胞浸润,将AQP5的表达上调,其机制可能与麦冬地芍汤上调血清及颌下腺中VIP的表达相关。
[Abstract]:Objective: to explore the theoretical basis of traditional Chinese medicine for the treatment of Sjogren's syndrome. Objective: to explore the treatment of Sjogren's syndrome (Sjogren's syndrome) with Radix Ophiopogonis decoction for regulating Th1/Th2. Th17/Treg immune imbalance and abnormal AQP5 expression were correlated with the up-regulation of neuropeptide VIP. Methods Thirty three female NOD mice were randomly divided into three groups: model group and hydroxychloroquine group. Eight ICR Institute of Cancer research mice in Chinese medicine group were divided into blank group and hydroxychloroquine group. The rats in the traditional Chinese medicine group were given hydroxychloroquine solution and Ophiopogonium root decoction respectively. The submandibular gland and serum were collected in the blank group and the model group at the age of 18 weeks. The saliva flow rate of the mice at the age of 101316 weeks was measured. The contents of VIP and cytokine IFN- 纬 IL-10 IL-17A in serum and one side of submandibular gland were detected by Elisa method, and HE staining was performed on the other side of submandibular gland. Results: 1. Compared with the blank group, the saliva flow rate of the model group was significantly lower than that of the model group. The saliva flow rate in the Chinese medicine group was significantly improved at the age of 16 weeks. The concentration of VIP in serum and submandibular gland of the traditional Chinese medicine group was significantly higher than that of the model group, and the level of IFN- 纬 in the serum and submandibular gland of the traditional Chinese medicine group was significantly higher than that of the model group. Compared with the model group, the level of IL-17A was significantly lower than that of the model group. The ratio of Thl / Th2T17 / Treg was significantly lower than that of the model group. The lymphocyte infiltration in submandibular gland of NOD mice was more obvious than that in normal mice, and was treated with Maidong Di Shao decoction. The pathological score of submandibular gland was improved significantly (P 0.05). The expression of AQP5 in the submandibular gland of the model group was significantly decreased by immunohistochemistry. The expression of AQP5 in hydroxychloroquine group and traditional Chinese medicine group was higher than that in model group P0.050.Conclusion: Meidongdi Shao decoction has significant curative effect on NOD mice model of Sjogren syndrome and improves saliva flow rate of mice. It regulated the immune balance of Thl / Th2 and Th17 / Treg, improved submandibular gland lymphocytic infiltration, and up-regulated the expression of AQP5. The mechanism may be related to the up-regulation of VIP expression in serum and submandibular gland by Maidong Dizhao decoction.
