艾灸穴位预处理增强大鼠胃黏膜防御功能机制的研究

【摘要】:目的:观察艾灸预处理大鼠足三里、中脘穴位,对无水乙醇灌胃造成的大鼠应激性胃黏膜损伤的影响,及大鼠胃组织EGFR(表皮生长因子受体)与大鼠血清中TGF-α(转化生长因子-α)含量的指标表达,探讨艾灸穴位预处理在增强大鼠胃黏膜防御功能方面的作用,为艾灸在防治胃黏膜损伤的疾病方面提供可靠的科学内涵及在临床中的可行性依据。材料与方法:健康SD大鼠60只,SPF级,体重200~250g±,随机分组为:A:空白组、B:模型组、C:艾灸穴位组、D:艾灸对照点组、E:奥美拉唑组,共5组,每组12只。实验前八天,空白组、模型组、奥美拉唑组大鼠正常饲养,每天予以与艾灸组相同的30min捆绑固定;艾灸穴位组大鼠将四肢及腹部用皮筋系于自制鼠板上固定,暴露施灸处穴位,艾灸足三里、中脘穴,每日30min;艾灸对照点组大鼠固定方法与艾灸组相同,艾灸足三里穴、中脘穴旁开的非穴位对照点。第八天各组大鼠按相应因素处理之后,禁食不禁水24小时,于造模前30min,空白组、模型组、艾灸穴位组、艾灸对照点组大鼠每只按2ml/100g的剂量腹腔注射生理盐水,奥美拉唑组大鼠每只按2ml/100g的剂量腹腔注射奥美拉唑;30min后,除空白组外,模型组、艾灸穴位组、艾灸对照点组、奥美拉唑组大鼠每只按0.5ml/100g的剂量灌胃无水乙醇,复制胃黏膜损伤模型;造模1h后,麻醉动物,采用激光散斑扫描影像系统观察大鼠胃血流量循环情况;肉眼观察大鼠胃黏膜的损伤程度,记录并进行损伤指数评分;光镜下观察胃黏膜组织形态的变化;酶联免疫吸附测定法(ELISA)检测大鼠血清中TGF-α的含量表达;免疫组织化学法检测胃组织中EGFR的表达。采用SPSS17.0统计软件分析并统计,样本以均数±标准差(x±s)表示,用单因素方差分析。结果:1.大鼠胃黏膜损伤指数(UI):与空白组相比,模型组胃黏膜损伤程度明显高于空白组(P0.01)其变化有统计学意义,说明造模成功;与模型组相比,艾灸对照点组胃黏膜损伤程度较模型组减轻(P0.05);与艾灸对照点组比较,艾灸穴位组、奥美拉唑组胃黏膜损伤指数有所降低(P0.05);与艾灸穴位组比较,奥美拉唑组的胃黏膜损伤指数较艾灸穴位组高(P0.01);说明艾灸穴位能减轻大鼠胃黏膜损伤的程度。2.大鼠胃血流量的表达:与空白组相比,模型组胃血流量低于空白组(P0.01);与模型组相比,艾灸穴位组、艾灸对照点组、奥美拉唑组胃血流量高于模型组(P0.01);与艾灸对照点组比较,艾灸穴位组、奥美拉唑组胃血流量较艾灸对照点组高(P0.01);与奥美拉唑组比较,艾灸穴位组胃血流量高于奥美拉唑组对比差异有统计学意义(P0.01);说明艾灸穴位能促进大鼠胃血流量的循环。3.大鼠胃黏膜组织形态光镜下观察:与空白组相比,模型组大鼠胃黏膜可见炎性细胞浸润,损伤明显;与模型组相比,艾灸穴位组、奥美拉唑组,胃黏膜表面的水肿,充血减少;艾灸对照点组胃黏膜有损伤及破损,但较模型组有所改善;与奥美拉唑组比较,艾灸穴位组胃黏膜破损改善相对明显,说明艾灸大鼠穴位,能保持无水乙醇损伤造成的大鼠胃黏膜组织形态的完整性。4.大鼠血清中TGF-α的含量表达:与空白组相比,模型组血清中TGF-α含量低于空白组(P0.01);与模型组相比,艾灸穴位组、艾灸对照点组、奥美拉唑组血清中TGF-α含量高于模型组(P0.05);与艾灸对照点组比较,艾灸穴位组、奥美拉唑组血清中TGF-α含量高于艾灸对照点组(P0.01);与艾灸穴位组比较,奥美拉唑组的血清中TGF-α含量较艾灸穴位组低,其差异有统计学意义,(P0.01);说明艾灸穴位能促进大鼠血清中TGF-α含量的表达。5.大鼠胃组织中EGFR的含量表达:与空白组相比,模型组胃组织中EGFR含量低于空白组(P0.01);与模型组相比,艾灸穴位组、艾灸对照点组、奥美拉唑组胃组织中EGFR含量高于模型组(P0.05);与艾灸对照点组比较,艾灸穴位组、奥美拉唑组胃组织中EGFR含量有所提高(P0.01);与艾灸穴位组比较,奥美拉唑组的胃组织中EGFR含量低于艾灸穴位组(P0.01),其差异有统计学意义;说明艾灸穴位能促进大鼠胃组织中EGFR含量的表达。结论:1.艾灸穴位预处理能降低大鼠胃黏膜损伤程度,提高了大鼠胃黏膜的防御功能。2.艾灸穴位预处理能调节大鼠胃血流量,增强大鼠胃的血流量循环,为预防大鼠胃黏膜损伤提供了条件。3.艾灸穴位预处理能改善光镜下大鼠胃黏膜组织形态。4.艾灸穴位预处理能提高大鼠血清中TGF-α的含量,促进了大鼠胃黏膜保护因子增殖,起到了保护胃黏膜的作用。5.艾灸穴位预处理能促进大鼠胃组织中EGFR的表达增强,增强抗大鼠胃黏膜的损伤能力。
[Abstract]:Objective: To observe the effect of moxibustion preconditioning in rats at Zusanli, Zhongwan acupoint, impact on ethanol gavage stress-induced gastric mucosal injury in rats, and rat gastric tissue EGFR (epidermal growth factor receptor) and TGF- in rat serum alpha (transforming growth factor alpha) expression content index, to study the effect of moxibustion on acupoint pretreatment in enhancing the defense function of gastric mucosa in rats the effect of moxibustion for feasibility basis in prevention and treatment of gastric mucosal injury disease and provide scientific connotation and reliable in clinical. Materials and methods: 60 healthy SD rats, weight SPF, 200~250g +, were randomly divided into five groups: A: control group. B: model C: group, Acupoint Moxibustion group, moxibustion group D:, E: omeprazole group, 5 group, 12 rats in each group. Eight days before the experiment, the blank group, model group, omeprazole group of rats fed normal every day to the same with the moxibustion group 30min moxibustion group is bound and fixed; In the limbs and abdominal elastic system in rat plate fixation, exposed moxibustion acupoints, moxibustion at Zusanli, Zhongwan, 30min daily; moxibustion group rats were fixed with the same method of moxibustion group, moxibustion at Zusanli, Zhongwan by non acupoint control group. After eighth days of each group rats were treated according to the corresponding factors, fasted for 24 hours, the 30min, the blank control group, model group, moxibustion group, moxibustion group rats per dose by intraperitoneal injection of saline 2ml/100g, omeprazole group rats each at a dose of 2ml/100g abdominal cavity injection of Omeprazole; after 30min. Except the blank group, model group, moxibustion group, moxibustion group, omeprazole group rats per gavage at a dose of 0.5ml/100g ethanol induced gastric mucosa injury model; animal model of 1h after anesthesia, using laser speckle image scanning system to observe the gastric blood of rats Flow circulation; the degree of injury observed in rat gastric mucosa, record and damage index; to observe the changes of morphology of gastric mucosal tissue under light microscope; enzyme-linked immunosorbent assay (ELISA) detection of TGF- expression levels in rat serum alpha; to detect the expression of EGFR in gastric tissue by immunohistochemical method. Using SPSS17.0 statistical analysis software and statistics, sample mean standard deviation (x + s), using single factor variance analysis. Results: the injury index of gastric mucosa of 1. rats (UI) compared with control group, the degree of gastric mucosal injury in model group was significantly higher than the control group (P0.01) the change was statistically the significance, indicating that the model was successful; compared with the model group, moxibustion group of gastric mucosa injury was slighter than that of model group (P0.05); compared with the control group of moxibustion, moxibustion group, omeprazole group, gastric mucosal injury index is lower (P0.05); and moxibustion group than in Relatively, gastric mucosal injury index of omeprazole group compared with moxibustion group (P0.01); high expression of moxibustion can reduce gastric mucosal injury in rats gastric blood flow level.2. rats: compared with the control group, model group, gastric blood flow was lower than that of the control group (P0.01); compared with the model group, Acupoint Moxibustion group moxibustion group, omeprazole group, gastric blood flow was higher than in the model group (P0.01); compared with the control group of moxibustion, moxibustion group, omeprazole group and gastric blood flow compared with moxibustion group high (P0.01); compared with omeprazole group, omeprazole group was higher than that of contrast group difference point moxibustion gastric blood flow was statistically significant (P0.01); that moxibustion can promote the gastric blood flow in rats of circulating.3. in gastric mucosa of rats with light microscope: compared with the blank group, the model group of rat gastric mucosa inflammatory cell infiltrate, injury significantly; compared with the model group, moxibustion Acupoint group, omeprazole group, gastric mucosal edema, congestion reduction; moxibustion at control of gastric mucosal damage and damage, but compared with the model group was improved; compared with omeprazole group, Acupoint Moxibustion group improved gastric mucosal damage is relatively obvious, that moxibustion in rats, the expression of TGF- in serum alpha integrity in.4. rats can keep caused by ethanol injury of rat gastric mucosa in morphology: compared with the control group, the content of serum alpha TGF- in the model group was lower than control group (P0.01); compared with the model group, moxibustion group, moxibustion group, omeprazole group in serum TGF- content was higher than that of the model group (P0.05); compared with the control group of moxibustion, moxibustion group, omeprazole TGF- alpha in serum was higher than moxibustion at control (P0.01); compared with moxibustion group, omeprazole group in serum alpha TGF- content than moxibustion acupoint group, the difference was Statistical significance (P0.01); that moxibustion can promote the expression of EGFR in TGF- alpha content in serum of rats gastric tissue.5. expression in rats: compared with the blank group, the content of EGFR in gastric tissue of model group was lower than control group (P0.01); compared with the model group, moxibustion group, moxibustion group the content of EGFR, omeprazole group in gastric tissue was higher than in the model group (P0.05); compared with the control group of moxibustion, moxibustion group, omeprazole group EGFR content in gastric tissue increased (P0.01); compared with moxibustion group, omeprazole group EGFR content in gastric tissue below the Acupoint Moxibustion group (P0.01), there was statistical significance the difference; that moxibustion can promote the expression of EGFR in gastric tissue in rats. Conclusion: 1. Acupoint Moxibustion preconditioning can reduce the damage degree of gastric mucosa of rats, improve the rat gastric mucosa defense function.2. Acupoint Moxibustion preconditioning can regulate rat Gastric blood flow, enhance blood flow circulation of rat stomach, to prevent gastric mucosal injury in rats with.3. Acupoint Moxibustion preconditioning can improve gastric mucosa of rats under light microscope morphology of.4. Acupoint Moxibustion pretreatment could improve the content of TGF- in serum of rats, promote the proliferation of rat gastric mucosa protection, played the role of.5. Acupoint Moxibustion pretreatment can promote the expression of EGFR in gastric tissue of rats in protecting gastric mucosa, enhance the ability of anti rat gastric mucosal injury.
