促肾上腺皮质激素释放激素2型受体基因多态性与调神健脾针刺法疗效相关性研究

【摘要】：目的:研究促肾上腺皮质激素释放激素2型受体相关单核苷酸序列的多态性与肠易激综合征调神健脾针刺法疗效的相关性。方法:将随机分入针刺组的116例患者进行为期6周的调神健脾针刺法治疗,第0周和第6周填写IBS-SSS量表作为症状等级评价的依据,归类分组为有效组和无效组。所有患者的血样经过QIAGEN法提取DNA进行DNA以及CRHR2相关单核苷酸链基因多态性的分析。通过互联网上搜索国际SNP数据库,检索出在中国人群中rs2284217、rs2284220和rs3779250中以A/G为表现型,rs2240403、rs2267710以及rs4722999中以C/T为表现型。筛选具有统计学差异的6条单核苷酸链rs4722999、rs3779250、rs2240403、rs2267710、rs2284217和rs2284220作为基因多态性分析对象。统计分析方法中DNA分析采用哈迪-温伯格平衡检验,CRHR2基因多态性与调神健脾针刺法疗效相关性采取3×2表卡方检验和四格表检验。结果:1.基线期有效组与无效组患者在年龄、体重、病程及IBS-SSS基线期分数上无统计学差异(P0.05),哈迪-温伯格检验结果示全部P0.05,说明群体基因遗传平衡,数据来自同一蒙德尔群体。2.rs2284217、rs2284220基因型AG与调神健脾针刺法疗效具有相关性,差异具有统计学意义(P0.05)。3.单核苷酸链rs2284217、rs2284220、rs3779250中的基因型AG与调神健脾针刺法疗效具有相关性,差异具有统计学意义(P0.05)。结论:肠易激综合征患者rs2284217、rs2284220、rs3779250上携带的基因型AG与调神健脾针刺法疗效具有相关性,采用调神健脾针刺法治疗携带该类基因分型患者的疗效明显优于其他CRHR2基因亚型患者。
[Abstract]:Objective: to study the relationship between the polymorphism of single nucleotide sequence associated with adrenocorticotropin releasing hormone type 2 receptor and the therapeutic effect of acupuncture of regulating spirit and strengthening spleen in irritable bowel syndrome. 116 patients in acupuncture group were randomly divided into acupuncture group and treated with acupuncture therapy of regulating spirit and strengthening spleen for 6 weeks. At week 0 and week 6, the IBS-SSS scale was filled out as the basis for the evaluation of symptom grade. All patients' blood samples were extracted from DNA by QIAGEN method for the analysis of DNA and CRHR2 related single nucleotide chain gene polymorphism. International SNP database. Rs2284217rs2284220 in Chinese population and rs2240403 in rs3779250 with A / G as phenotype were obtained. Using C / T as phenotype in rs2267710 and rs4722999, six single nucleotide chains rs4722999 and rs3779250 with statistical difference were screened. Rs2240403,rs2267710. Rs2284217 and rs2284220 were used as gene polymorphism analysis objects. Hardy Weinberg equilibrium test was used for DNA analysis in statistical analysis method. The correlation between the polymorphism of CRHR2 gene and the therapeutic effect of acupuncture of regulating spirit and strengthening spleen was determined by using 3 脳 2 table chi-square test and four-grid test. Results: 1. The age and weight of the patients in the baseline effective group and the ineffective group were in age and weight. There was no statistical difference in the course of disease and the score of IBS-SSS baseline period. The results of Hardy Weinberg test showed that all of them were P0.05, indicating the genetic balance of the population. Data from the same Mondel population. 2.rs2284217rs2284220 genotype AG was correlated with the therapeutic effect of acupuncture for regulating spirit and invigorating spleen. The difference was statistically significant (P 0.05). Single nucleotide chain rs2284217rs2284220. The genotype AG in rs3779250 was correlated with the acupuncture effect of regulating spirit and invigorating spleen, and the difference was statistically significant (P 0.05). Conclusion: the patients with irritable bowel syndrome have rs2284217. The genotype AG carried on rs2284220rs3779250 was correlated with the therapeutic effect of acupuncture of regulating spirit and strengthening spleen. The therapeutic effect of Tiaoshen Jianpi acupuncture was better than that of other subtypes of CRHR2 gene in the treatment of patients with this kind of genotyping.
