中药中空栓对围绝经期综合征性激素及β-内啡肽影响的实验研究

【摘要】:目的:通过制备中药中空栓(复方地萸中空栓)作用于围绝经期综合征大鼠模型后检测相关指标,从分子水平探讨复方地萸中空栓对围绝经期综合征的影响及作用机制,为进一步研究及临床使用奠定基础。方法:1取复方中熟地、山茱萸、黄连、丹参、钩藤、香附、山药、茯神、远志、百合、葛根、浮小麦、甘草共13味药的免煎颗粒放入研钵内进行研磨,过120目筛,得复方免煎颗粒药粉,通过文献、导师前期研究成果及相关实验确定的具体制备方法,制备出大鼠用中药中空栓每粒约重1.905g,含复方免煎颗粒药量0.333g。1选48只生育龄雌性未孕SD大鼠,随机分成假手术组、模型组、阳性组、中药灌胃组、中药中空栓高剂量组、中药中空栓低剂量组。除假手术组背部切口切除少量脂肪组织后立即缝合切口皮肤外,其余5组大鼠手术切除双侧卵巢,制备围绝经期综合征大鼠模型。各组大鼠术后恢复性喂养一周,并予前三天连续肌肉注射青霉素4万个单位,1次/天,以防感染。一周后,各组大鼠进行阴道细胞学涂片观察,1次/天,连续五天,剔除去势不彻底(造模不成功)的大鼠。1阴道涂片检查后,假手术组及模型组给予空白中空栓,阳性组给予戊酸雌二醇中空栓,中药灌胃组灌服复方地萸混悬液,中药中空栓高剂量组与低剂量组分别给予相应剂量的复方地萸中空栓。连续给药14天,末次给药后24小时准备杀检,杀检前12个小时禁食不禁水,腹主动脉取血,迅速断头处死大鼠,摘取下丘脑。用Elisa试剂盒测定血清性激素雌二醇(E_2)、卵泡刺激素(FSH)、黄体生成素(LH)及下丘脑β-内啡肽(β-EP)。结果:1复方地萸中空栓能够提高大鼠血清E_2水平的同时降低FSH、LH的水平,较模型组有显著差异(P0.05),较阳性组、中药灌胃组无显著差异(P0.05);1复方地萸中空栓能够提高下丘脑β-EP含量,较模型组有显著差异(P0.05),较阳性组、中药灌胃组无显著差异(P0.05)。结论:复方地萸中空栓能调节围绝经综合征大鼠模型血清性激素水平及下丘脑β-EP的分泌,使E_2、β-EP升高,FSH、LH降低,从而改善围绝经期综合征的临床症状。其机制可能是通过调节这些指标直接或间接参与调节围绝经期综合征神经内分泌系统功能。
[Abstract]:Objective: to detect the related indexes after the preparation of traditional Chinese medicine hollow suppository (compound Diyu hollow suppository) on the model of peri-menopausal syndrome in rats. To explore the effect and mechanism of compound Diyu hollow suppository on peri-menopausal syndrome from molecular level, and to lay a foundation for further study and clinical application. Methods: 1. Take compound Corni officinalis, Fructus Corni, Coptis chinensis and Salvia miltiorrhiza (Salvia miltiorrhiza). Uncaria, fragrant, yam, Fu Shen, Yuan Zhi, Lily, Pueraria root, floating wheat, licorice a total of 13 kinds of medicine granules into the mortar for grinding, after 120 mesh sieve, obtained compound Fan-Fen granule powder, through the literature. The former research results and the specific preparation methods determined by related experiments, the preparation of Chinese medicine hollow suppository for rats weighs about 1.905g. 48 female SD rats with reproductive age were randomly divided into sham-operation group, model group, positive group, Chinese medicine gavage group and high dose group of Chinese medicine hollow suppository. The low-dose group of hollow suppository: except for the sham operation group, the skin of the incision was sutured immediately after a small amount of fat tissue was removed from the back of the sham operation group, and the other 5 groups of rats were operated to remove bilateral ovaries. The model of rats with peri-menopausal syndrome was established. The rats in each group were fed for one week after operation, and penicillin was injected intramuscularly once a day for three days to prevent infection. The vaginal cytologic smears were observed once a day for five consecutive days. After removing the castrated rats, the sham operation group and the model group were given blank hollow suppository. The positive group was given estradiol valerate hollow suppository, the Chinese medicine gavage group was infused with compound Diyu suspension, the high dose group and the low dose group of Chinese medicine hollow suppository were given the corresponding dose of compound diyu hollow suppository for 14 days. The rats were killed 24 hours after the last administration. The rats were killed by fasting 12 hours before the last administration. Blood was taken from the abdominal aorta and the rats were killed quickly. Hypothalamus was removed. Serum estradiol estradiol (E _ 2) and follicle stimulating hormone (FSH) were determined by Elisa kit. Results the hypothalamus 尾 -endorphin (尾 -EP) and luteinizing hormone (LH) could increase the serum E2 level and decrease the FSHLH level in rats. Compared with the model group, there was a significant difference between the two groups, but there was no significant difference between the Chinese medicine group and the positive group. 1 compound Diyu hollow suppository could increase 尾 -EP content in hypothalamus, which was significantly different from model group (P 0.05) and positive group (P < 0.05). Conclusion: compound Diyu hollow suppository can regulate the level of serum sex hormone and the secretion of 尾 -EP in hypothalamus of rats with peri-menopausal syndrome. 尾 -EP increased and FSH LH decreased. Therefore, the clinical symptoms of peri-menopausal syndrome can be improved. The mechanism may be to regulate the neuroendocrine system function of perimenopausal syndrome directly or indirectly by regulating these indexes.
