针灸预处理对成年和老年福尔马林致痛大鼠脊髓NO分布和c-fos表达的影响

【摘要】:目的比较成年和老年大鼠对炎症性疼痛反应的差别,及针灸预处理对成年和老年福尔马林致痛大鼠脊髓一氧化氮(Nitric oxide,NO)分布和c-fos表达的影响。方法将4月龄和21月龄雄性Sprague Dawley(SD)大鼠,分别随机分为对照组(不做任何处理)、福尔马林组(仅注射福尔马林)、针灸足三里穴+福尔马林组(针灸足三里穴后注射福尔马林)、针灸非穴位+福尔马林组(针灸非穴位后注射福尔马林),每组6只大鼠。针灸处理后,除对照组外,其余各组于针灸位点同侧足底注射福尔马林。观察大鼠行为学反应,记录福尔马林注射后1小时内的缩足次数和舔/咬足时间(秒),1.5小时后灌流取脊髓第4腰髓(4th Lumbar spinal cord,L4)至第5腰髓(5th Lumbar spinal cord,L5)节段,采用酶组织化学染色和免疫组织化学染色法检测脊髓NO的分布和c-fos的表达。结果1、对照组:NO数量较低,且老年大鼠脊髓背角NO数量低于成年大鼠(P0.01);Fos免疫阳性神经元基础水平较低,且在成年和老年大鼠间无差异(P0.05);成年和老年大鼠脊髓未发现NO和Fos双标阳性神经元。2、福尔马林组:老年大鼠继发疼痛时相的缩足次数高于成年大鼠(P0.05);与对照组比较,NO阳性神经元数、Fos免疫阳性神经元数和二者双标阳性神经元数在成年和老年大鼠脊髓均增加(P0.01),且老年大鼠的NO阳性神经元数(P0.05,脊髓背角;P0.01,中央管)和Fos免疫阳性神经元数(P0.01)高于成年大鼠。3、针灸足三里穴+福尔马林组:成年和老年大鼠继发疼痛时相的缩足次数(P0.01)和舔/咬足时间(成年,P0.05;老年,P0.01)低于福尔马林组;成年和老年大鼠脊髓NO阳性神经元数、Fos免疫阳性神经元数和二者双标阳性神经元数均低于福尔马林组(P0.01);且老年组NO阳性神经元数和Fos免疫阳性神经元较福尔马林组下降的幅度高于成年组大鼠(P0.01)。4、针灸非穴位+福尔马林组:成年和老年大鼠继发疼痛时相的缩足次数、舔/咬足时间、NO阳性神经元数、Fos免疫阳性神经元数和二者双标阳性神经元数与福尔马林组比较无差异(P0.05)。结论老年大鼠对炎症性疼痛的反应强于成年大鼠,针灸预处理足三里穴可以提高大鼠痛阈,减轻疼痛反应,降低脊髓NO的数量和c-fos的表达,且对老年大鼠的作用优于成年大鼠。
[Abstract]:Objective to compare the response of adult and aged rats to inflammatory pain. The effects of acupuncture and moxibustion preconditioning on the distribution of nitric oxide nitric oxide (no) and the expression of c-fos in spinal cord of adult and aged formalin-induced pain rats were studied. They were randomly divided into the control group (without any treatment), the formalin group (only injected with formalin, the acupuncture and moxibustion Zusanli point formalin group (acupuncture and moxibustion at Zusanli point after injection of formalin), the acupuncture and moxibustion non-acupoint formalin group (Acupuncture and moxibustion non-acupoint formalin group). After acupuncture and moxibustion, 6 rats in each group were injected with formalin after acupuncture and moxibustion. After acupuncture and moxibustion treatment, Except for the control group, the other groups were injected with formalin on the ipsilateral plantar of the acupuncture and moxibustion site. The number of foot contractions and the time of licking / biting were recorded within 1 hour after formalin injection (the 4th Lumbar spinal cordlordant L4 of the 4th Lumbar spinal of the spinal cord was perfused after 1.5 hours) to the 5th Lumbar spinal cordlord-L5 segment of the spinal cord. Enzyme histochemical staining and immunohistochemical staining were used to detect the distribution of no and c-fos expression in spinal cord. The basic level of Fos immunoreactive neurons in the spinal dorsal horn of aged rats was lower than that in adult rats. There was no difference between adult rats and old rats (P 0.05); no and Fos double labeled positive neurons were found in the spinal cord of adult and old rats. In formalin group, the number of foot contraction in the secondary pain phase of the aged rats was higher than that in the adult rats, and compared with the control group. The number of Fos immunoreactive neurons and the number of double labeled neurons increased in the spinal cord of adult and aged rats, and the number of no positive neurons in aged rats was P0.05, P0.01 in dorsal horn of spinal cord, central canal) and Fos immunized positive neurons. The number of sex neurons (P0.01) was higher than that of adult rats (P0.01), and the number of feet contraction (P0.01) and the time of licking / biting (P0.05; P0.01) were lower in the acupuncture and moxibustion Zusanli (Zusanli) group than in the formalin group. The number of Fos immunoreactive neurons and the number of double labeled neurons in spinal cord of adult and aged rats were lower than those of formalin group (P 0.01), and the number of no positive neurons and Fos immunoreactive neurons in the aged group was higher than that in formalin group. The decrease in the group was higher than that in the adult group (P 0.01), and that in the acupuncture and moxibustion non-acupoint formalin group: the number of foot contractions in the stage of secondary pain in adult and aged rats, There was no difference in the number of Fos immunoreactive neurons and the number of double labeled neurons between the two groups compared with formalin group. Conclusion the response to inflammatory pain in aged rats is stronger than that in adult rats. Acupuncture and moxibustion pretreatment at Zusanli point could increase the pain threshold, alleviate the pain response, decrease the number of no and c-fos expression in spinal cord, and the effect on aged rats was better than that of adult rats.
