咳喘宁对RSV诱发哮喘大鼠血清IL-4、IFN-γ和IgE的调节作用

【摘要】:目的:观察咳喘宁对RSV诱发哮喘大鼠血清IL-4、IFN-γ和IgE表达的影响,探讨其防治哮喘的机制。方法:将60只雄性幼年大鼠随机分为6组,即正常组,模型组,泼尼松与沙丁胺醇治疗组(简称西药治疗组),咳喘宁大、中、小剂量组,每组各10只。除正常组外,其余五组大鼠注射由PBS稀释的1ml无菌抗原液(含10mg卵清白蛋白+100 mg氢氧化铝凝胶)致敏;以1%卵清蛋白生理盐水溶液雾化诱喘;鼻腔滴入RSV激发哮喘。正常组以生理盐水进行雾化及滴鼻。造模完成当天进行治疗:西药治疗组予混悬液醋酸泼尼松、沙丁胺醇片剂,咳喘宁大、中、小剂量组予大中小剂量的咳喘宁治疗,正常组及模型组予蒸馏水灌胃,每日灌胃1次。连续治疗7天后,取腹主动脉血,分离血清。用ELISA进行IL-4、IFN-γ、IgE水平的测定。结果:?与正常组比较,哮喘组血清IL-4、IgE值明显升高(P0.01),IFN-γ值明显降低(P0.01);与模型组相比,西药治疗组、咳喘宁各剂量组血清IL-4、IgE值明显降低(P0.05),而IFN-γ明显升高(P0.05);与西药治疗组比较,咳喘宁中、小剂量组血清IL-4、IgE值无明显变化(P0.05),咳喘宁各剂量组血清IFN-γ值无明显变化(P0.05);与大、小剂量组相比,中剂量组血清IL-4、IgE值降低较为明显(P0.05),中剂量组血清IFN-γ值相对升高(P0.05)。?检测血清IgE水平与IL-4水平相关性,示IgE与IL-4水平呈正性强相关(r=0.628,P值0.01);检测血清IgE水平与IFN-γ水平相关性,示IgE水平与IFN-γ水平呈负性中等程度相关(r=-0.591,P值0.01)。结论:咳喘宁治疗RSV诱发哮喘大鼠的作用机制之一可能是通过上调IFN-r水平、降低IL-4和IgE水平表达从而降低气道炎症反应,维持Th1/Th2平衡来调节机体免疫机制。
[Abstract]:Objective: to observe the effect of Kechuanning on the expression of IL-4, IFN- 纬 and IgE in serum of asthmatic rats induced by RSV, and to explore its mechanism of preventing and treating asthma. Methods: 60 young male rats were randomly divided into 6 groups: normal group and model group. Prednisone and salbutamol treatment group (referred to as western medicine treatment group, Kechuanning large, medium and small dosage groups, each group, 10 rats, except normal group, The other five groups were sensitized by PBS diluted 1ml aseptic antigen solution (containing 10mg ovalbumin 100mg aluminum hydroxide gel) and inhaled with 1% ovalbumin saline solution. The normal group was treated with normal saline on the day of nebulization and nasal drip: the western medicine treatment group was treated with prednisone acetate, salbutamol tablet, Kechuanning large, medium, the western medicine treatment group was given the suspension of prednisone acetate, salbutamol tablet, Small dose group was treated with Kechuanning, normal group and model group were fed with distilled water once a day. After 7 days of continuous treatment, abdominal aorta blood was taken and serum was separated. IL-4 IFN- 纬 -IgE level was measured by ELISA. Compared with the normal group, the serum IL-4 IgE level in asthma group was significantly higher than that in the control group, and the level of IFN- 纬 was significantly lower in the asthmatic group than in the model group, compared with the model group, the serum IL-4 IgE level in the western medicine treatment group was significantly lower than that in the Kechuanning group, while the level of IFN- 纬 in the western medicine group was significantly higher than that in the western medicine treatment group, and in Kechuanning group, there was no significant difference between Kechuanning group and western medicine group. There was no significant change in serum IL-4 IgE value in low dose group and no significant change in serum IFN- 纬 value in Kechuanning group. Compared with large and low dose group, the serum IL-4 titer IgE value in middle dose group was significantly lower than that in medium dose group (P 0.05), and the serum IFN- 纬 value in medium dose group was relatively higher than that in medium dose group. The correlation between serum IgE level and IL-4 level showed that there was a positive correlation between IgE and IL-4 level (P = 0.01), and serum IgE level was correlated with IFN- 纬 level. The results showed that the level of IgE was negatively correlated with the level of IFN- 纬. Conclusion: one of the mechanisms of Kechuanning in the treatment of asthmatic rats induced by RSV may be to decrease the expression of IL-4 and IgE by upregulating the level of IFN-r and reducing the expression of IL-4 and IgE. Maintain the balance of Th1/Th2 to regulate the immune mechanism of the body.
