隔盐隔姜灸“神阙”穴对逼尿肌反射亢进大鼠尿动力及逼尿肌M2、M3受体蛋白表达的影响

【摘要】:目的:探讨隔盐隔姜灸"神阙"穴对逼尿肌反射亢进的效应及其作用机制。方法:成年雌性SD大鼠30只,其中20只大鼠采用T9完全脊髓横断方法建立逼尿肌反射亢进模型后随机分为模型组(10只)和艾灸组(10只),余10只作为假手术组行假脊髓横断手术。艾灸组大鼠出现尿失禁(约造模2周)后,给以隔盐隔姜灸"神阙"穴治疗,每次灸3壮,每日1次,连续治疗7d。各组在干预结束后,进行尿流动力学检测,之后取膀胱逼尿肌,采用免疫蛋白印迹法检测M2、M3受体蛋白表达情况。结果:与假手术组大鼠相比,模型组大鼠排尿间期明显较短(P0.01),最大膀胱内压明显较高(P0.01),膀胱逼尿肌M2受体蛋白表达显著升高(P0.05),M3受体蛋白表达无明显变化(P0.05);与模型组相比,艾灸组大鼠排尿间期明显较长(P0.01),最大膀胱内压明显降低(P0.01),膀胱逼尿肌M2受体蛋白表达显著下降(P0.05),而M3受体蛋白表达无明显变化(P0.05);与假手术组相比,艾灸组以上指标差异均无统计学意义(均P0.05)。结论:隔盐隔姜灸"神阙"穴能抑制脊髓横断大鼠的膀胱过度活动状态,其作用机制可能与减少膀胱逼尿肌M2受体蛋白的表达、抑制逼尿肌的过度收缩有关。
[Abstract]:Objective: to investigate the effect and mechanism of salt-separated ginger-separated moxibustion on detrusor hyperreactivity in 30 adult female SD rats. Twenty rats were randomly divided into model group (n = 10) and moxibustion group (n = 10). The remaining 10 rats were used as sham operation group for pseudo spinal cord transection. After incontinence (about 2 weeks), The ginger-separated moxibustion was treated with "Shenque" point, 3 strong moxibustion every time, once a day, continuously for 7 days. After the intervention, the urinary fluid dynamics was measured, and the bladder detrusor muscle was taken out. The expression of M2M3 receptor protein was detected by Western blot. Results: compared with sham-operated rats, the expression of M2M3 receptor protein was detected by Western blot. In the model group, the urinary interval was significantly shorter, the maximal intravesical pressure was significantly higher than that in the control group, and the expression of M2 receptor protein in the bladder detrusor muscle was significantly higher than that in the model group, and there was no significant change in the expression of P0.05M3 receptor protein in the bladder detrusor muscle compared with the model group. In the moxibustion group, the urination interval was significantly longer than that in the control group, the maximal intravesical pressure decreased significantly, and the expression of M2 receptor protein in the bladder detrusor muscle decreased significantly, while the expression of M3 receptor protein did not change significantly in the moxibustion group, compared with the sham-operated group, the expression of M2 receptor protein in the bladder detrusor muscle decreased significantly, and the expression of M3 receptor protein did not change in the moxibustion group. Conclusion: moxibustion with ginger-separated moxibustion on "Shenque" can inhibit the overactive state of bladder in rats with spinal cord transection, and its mechanism may be related to reducing the expression of M2 receptor protein in bladder detrusor muscle.
