“形神共养”对大鼠认知功能、突触素、微管相关蛋白-2及锥体细胞的影响

【摘要】:目的:探讨"形神共养"的丰富生存环境对正常大鼠认知功能、突触素(SYN)、微管相关蛋白-2(MAP-2)的表达及大脑m PFC区锥体细胞的影响。方法:健康雄性SD大鼠48只,随机分为"形养"组(EE1),"神养"组(EE2),"形神共养"组(EE3)和标准养组(SE),每组12只,分别进行形养、神养、形神共养的丰富生存环境训练及标准养。训练结束后进行水迷宫实验,水迷宫结束后用免疫组织化学方法检测海马区SYN、MAP-2的表达;用Golgi-Cox染色观察大脑内侧前额叶区锥体细胞树突的变化。结果:水迷宫测试的第5天,EE3组大鼠的潜伏期明显短于SE组(P0.05);4组大鼠在目标象限停留的时间无明显差异(P0.05)。SE组的SYN表达最低,而EE1组和EE3组之间的表达无明显差异且均高于EE2组。SE组的MAP-2表达最低,而EE3组的表达高于EE1组和EE2组。m PFC区锥体细胞树突交点总数4组组间比较差异无显著性意义(P0.05)。棘密度EE3组最大,其次是EE1组,SE组与EE2组之间无区别且较EE1和EE3组低。结论:"形神共养"的丰富生存环境可提高正常大鼠的认知功能,并促进SYN及MAP-2免疫表达及锥体细胞树突生长。
[Abstract]:Objective: to investigate the effects of the rich living environment of "form and spirit co-nourishment" on cognitive function, synaptophysin and microtubulosis-associated protein (-2mMAP-2) expression in normal rats and the pyramidal cells in the m PFC region of the brain. Methods: 48 healthy male Sprague-Dawley rats were included in this study. They were randomly divided into three groups: "form nourishing" group, "spirit nourishing" group, "form and spirit common nourishing" group, "form and spirit raising together" group, and standard group, 12 rats in each group were trained in rich living environment and standard culture. After training, water labyrinth experiment was carried out. After the water maze was finished, the expression of SYN-MAP-2 in hippocampal area was detected by immunohistochemical method. The changes of dendritic cells of pyramidal cells in the medial prefrontal lobe of the brain were observed by Golgi-Cox staining. Results: the latency of rats in the EE3 group on the 5th day of water maze test was significantly shorter than that of the rats in the target quadrant of the SE group P0.05 + 4. There was no significant difference in the retention time of the rats in the target quadrant. The expression of SYN was the lowest in the group, However, there was no significant difference between EE1 group and EE3 group and the lowest expression of MAP-2 was found in EE2 group and SE group. However, the expression of dendritic cells in EE3 group was higher than that in EE1 group and EE2 group. There was no significant difference in the total number of dendritic cells in pyramidal cell in EE2 group. There was no significant difference between four groups (P 0.05). The spike density was the highest in EE3 group. Secondly, there was no difference between SE group and EE2 group in EE1 group, which was lower than that in EE1 and EE3 group. Conclusion: the rich living environment of "form and spirit co-nourishing" can improve the cognitive function of normal rats, and promote the expression of SYN and MAP-2 and the dendritic growth of pyramidal cells.
