针刺对血管性痴呆大鼠海马Ref-1上下游调控分子的影响

【摘要】:目的:观察针刺对血管性痴呆大鼠海马Ref-1上下游调控分子的影响,探讨针刺基于Ref-1信号通路的抗氧化机制。方法:Wistar大鼠82只,随机分为假手术组、模型组、针刺组和非穴组。双侧颈总动脉阻断法复制血管性痴呆大鼠模型,造模后第3天针刺,针刺治疗结束后,采用Morris水迷宫检测大鼠认知功能;尼氏染色检测大鼠海马神经元凋亡情况;Western blot技术检测大鼠海马磷酸化CKⅡ蛋白表达水平;免疫组化技术检测大鼠海马HIF-1α蛋白表达情况。结果:与假手术组比较,造模后大鼠认知功能受损,海马区神经元凋亡,磷酸化CKⅡ水平显著下降(P0.05),HIF-1α表达水平显著升高(P0.05)。针刺组大鼠学习记忆能力及神经元凋亡情况显著改善,磷酸化CKⅡ水平显著升高(P0.05),HIF-1α表达水平显著降低(P0.05),且改善程度显著优于非穴组(P0.05)。结论:针刺通过提高海马Ref-1上游CKⅡ的磷酸化表达水平,降低海马Ref-1下游HIF-1α表达水平,发挥抗氧化作用,抑制神经元细胞凋亡,改善学习记忆能力。
[Abstract]:Objective: to observe the effect of acupuncture on the upstream and downstream regulation molecules of Ref-1 in hippocampus of vascular dementia rats, and to explore the antioxidant mechanism of acupuncture based on Ref-1 signaling pathway. Methods: 82 Wistar rats were randomly divided into sham operation group and model group. The rat model of vascular dementia was established by bilateral common carotid artery occlusion. After acupuncture on the 3rd day after acupuncture, the cognitive function of the rats was measured by Morris water maze. The apoptosis of hippocampal neurons in rats was detected by Nissl staining.
