针刺对MCAO大鼠脑功能恢复及p38MAPK,p-p38MAPK,GLT-1表达影响的研究

【摘要】:目的:制备大脑中动脉栓塞(MCAO)大鼠模型,通过针刺其内关穴,检测p38MA PK,p-p38MAPK,GLT-1在脑组织中的表达情况,初步探讨p38MAPK,p-p38MAPK,GLT-1对谷氨酸-谷氨酰胺循环的调控作用,为针刺治疗缺血性脑中风的作用机制研究提供新的研究思路和实验依据。方法:采用线栓法建立大脑中脉栓塞(MCAO)大鼠模型,将大鼠分为模型组、内关组和地机组,另取假手术组为实验对照组,内关组、地机组大鼠分别连续针刺内关穴、地机穴7d,对各组大鼠进行神经体征评分,评估神经功能恢复情况。多聚甲醛灌注后取脑,通过观察脑组织石蜡切片HE染色检测神经元梗死面积,免疫组织化学检测p38MAPK、p-p38MAPK、GLT-1表达变化;脑组织匀浆,利用Western Blot检测方法,观察针刺对MCAO大鼠脑组织p38MAPK、p-p38MAPK、GLT-1表达的影响。结果:1.连续针刺内关穴7d可显著改善MCAO大鼠神经功能,能够促进其运动功能的恢复,内关穴的效果明显优于地机穴。2.针刺内关穴显著上调MCAO大鼠脑组织中GLT-1的表达,下调p-p38MAPK、p38MAPK的表达,内关穴在影响p-p38MAPK、p38MAPK、GLT-1表达方面明显优于地机穴。结论:针刺内关穴能够促进GLT-1的表达,减少p-p38MAPK、p38MAPK的表达,从而能够显著降低缺血脑组织中过度堆积的谷氨酸浓度,减轻其神经毒性作用,有利于缺血脑组织神经功能的恢复。
[Abstract]:Objective: to establish a rat model of middle cerebral artery embolization (MCAO) and to detect the expression of p38 MAPK-1 GLT-1 in brain tissue by acupuncture at its Neiguan point, and to explore the regulatory effect of p38 MAPK- p-p38 MAPK- GLT-1 on glutamic acid-glutamine circulation.To provide new ideas and experimental basis for the study of the mechanism of acupuncture in the treatment of ischemic stroke.Methods: the MCAO rat model was established by thread embolization. The rats were divided into three groups: model group, Neiguan group and ground unit. The sham operation group was used as the experimental control group, the Neiguan group and the ground unit group were used as the experimental control group, the Neiguan group and the ground unit group respectively.After 7 days, the nerve signs were scored to assess the recovery of nerve function.The infarct size of neurons was detected by HE staining in paraffin sections of brain tissue, the expression of p38MAPKnp-p38MAPKngt GLT-1 was detected by immunohistochemistry, and the brain homogenate was detected by Western Blot method.To observe the effect of acupuncture on the expression of GLT-1 in brain tissue of MCAO rats.The result is 1: 1.Continuous acupuncture at Neiguan acupoint for 7 days could significantly improve the nerve function and promote the recovery of motor function in MCAO rats. The effect of Neiguan acupoint was better than that of Diji acupoint. 2.Acupuncture at Neiguan significantly upregulated the expression of GLT-1 and down-regulated the expression of p-p38 MAPK p38 MAPK in the brain of MCAO rats. Neiguan acupoint was superior to Diji acupoint in influencing the expression of p-p38 MAPK- p38 MAPK- GLT-1.Conclusion: acupuncture at Neiguan point can promote the expression of GLT-1 and decrease the expression of p-p38 MAPK- p38 MAPK, which can significantly reduce the excessive accumulation of glutamate in ischemic brain tissue, alleviate its neurotoxic effect, and be beneficial to the recovery of neural function in ischemic brain tissue.
