枫杨对类风湿关节炎成纤维滑膜细胞的增殖和凋亡的影响

【摘要】:目的:研究枫杨对类风湿关节炎成纤维滑膜细胞的作用,证实枫杨祛风除湿、消肿止痛的功效,为治疗痹症发掘新药。方法:制备枫杨萃取物的药液,用不同时间、不同浓度的药液刺激类风湿关节炎成纤维滑膜细胞。用CCK-8法、流式细胞术检测枫杨萃取物对类风湿关节炎成纤维滑膜细胞起到的作用。Western-Blot检测枫杨萃取物培养的类风湿关节炎成纤维滑膜细胞的caspase-3和caspase-8蛋白的表达。ELISA检测枫杨萃取物培养的类风湿关节炎成纤维滑膜细胞上清中IL-8的含量。结果:用不同浓度枫杨萃取物干预类风湿关节炎成纤维滑膜细胞,发现枫杨能够抑制细胞增殖,且呈时间和剂量依赖性(P0.05)。采用流式细胞仪检测枫杨萃取物诱导类风湿关节炎成纤维滑膜细胞凋亡情况,发现该药物能够有效诱导细胞凋亡(P0.01),2mg/ml药物浓度时凋亡细胞率为19.59%,当药物浓度从2.5mg/ml升高至3.33mg/ml时,细胞总凋亡率从42.03%增加到78.42%。用不同浓度枫杨萃取物培养的类风湿关节炎成纤维滑膜细胞,高剂量浓度组caspase-3和caspase-8蛋白表达上调(P0.05)。1.25mg/ml药物浓度时枫杨萃取物培养的类风湿关节炎成纤维滑膜细胞上清中IL-8的含量明显降低,且呈时间依赖性(P0.05)。结论:枫杨能抑制类风湿关节炎成纤维滑膜细胞的增殖,诱导类风湿关节炎成纤维滑膜细胞凋亡,并降低IL-8生成。有必要对其机制及其对类风湿关节炎治疗的临床效果做进一步研究。
[Abstract]:Objective: To study the effect of maple poplar on rheumatoid arthritis fibrosynovial cells, verify the effect of maple poplar to dispel wind and dehumidification, eliminate swelling and relieve pain, explore new medicine for the treatment of arthralgia syndrome. Method: preparation of the liquid of maple poplar extract, using different time and different concentration of liquid to stimulate rheumatoid arthrosis fibroblast synovial cells. CCK-8 method and flow cytometry are used. The effect of maple poplar extract on rheumatoid arthritis fibroblast synovial cells.Western-Blot detection of Caspase-3 and caspase-8 protein expression of rheumatoid arthritis fibroblast synovial cells cultured by Maple Yang Cui extract.ELISA detection of IL-8 content in the supernatant of rheumatoid arthritis fibroblast synovial cells cultured by Maple poplar extract. Fruit: using different concentrations of maple poplar extract to interfere with the fibroblast synovial cells of rheumatoid arthritis, it was found that maple poplar could inhibit cell proliferation and was time and dose dependent (P0.05). Flow cytometry was used to detect the withering of rheumatoid arthritis fibrosynovial cells induced by Maple poplar extract, and it was found that the drug could effectively induce cell withering. Death (P0.01), the apoptotic cell rate of 2mg/ml drug concentration was 19.59%. When the drug concentration increased from 2.5mg/ml to 3.33mg/ml, the total cell apoptosis rate increased from 42.03% to 78.42%. with different concentrations of maple poplar extract for rheumatoid arthritis fibroblast synovial cells, and the high dose concentration group caspase-3 and caspase-8 protein expression up (P0.05).1.25mg/ The content of IL-8 in the supernatant of rheumatoid arthritis fibrosynovial cells cultured by poplar extract at mL concentration significantly decreased and was time dependent (P0.05). Conclusion: Maple poplar can inhibit the proliferation of fibrous synovial cells of rheumatoid arthritis, induce apoptosis of fibroblast synovial cells in rheumatoid arthritis and reduce the formation of IL-8. It is necessary to Its mechanism and its clinical effect in the treatment of rheumatoid arthritis are further studied.
