三叶青藤木脂素苷类成分对小鼠胶原性关节炎的干预作用及对Th17细胞的调控作用研究

【摘要】:目的:探讨三叶青藤木脂素苷(LGIR)类成分对小鼠胶原性关节炎的治疗作用及对Th17细胞的增殖调控作用及其机制。方法:用DBA/1小鼠建立CIA小鼠模型,将造模成功的DBA/1小鼠平均分为CIA模型组、甲氨蝶呤(MTX)组、LGIR低、中、高剂量组,分别行药物灌胃干预,另设正常对照组以生理盐水干预。干预结束后检测各组小鼠血清IL-23和IL-17水平,观察各组小鼠踝关节滑膜病理改变,取脾脏并提取淋巴细胞,应用流式细胞术(Flow Cytometry,FCM)检测Th17细胞在不同组CIA小鼠淋巴细胞中的表达。结果:1.成功复制CIA小鼠模型。2.与模型组相比较,LGIR可改善CIA小鼠的AI评分。3.与模型组相比,LGIR可改善CIA小鼠踝关节病理评分。4.与模型组相比,LGIR可下调血清IL-23和IL-17水平。5.Th17淋巴细胞亚群比率的改变:模型组与正常对照组比较表现为Th17细胞亚群比例增加,LGIR高剂量组、MTX组与CIA模型组比较Th17细胞比例明显降低。结论:LGIR能够改善CIA小鼠局部关节肿胀程度,下调血清IL-23和IL-17水平,减轻关节病理改变程度,LGIR高剂量组可以下调CIA小鼠淋巴细胞中Th17细胞亚群比例。
[Abstract]:Aim: to investigate the therapeutic effect of three Ye Qing lividin glycosides on collagen-induced arthritis in mice and its mechanism in regulating the proliferation of Th17 cells. Methods: CIA mouse model was established with DBA / 1 mice. The successful DBA / 1 mice were divided into CIA model group, methotrexate MTX group and low, middle and high dose groups respectively. After the intervention, the levels of IL-23 and IL-17 in serum of each group were measured, the pathological changes of synovial membrane of ankle joint of each group were observed, the spleen was taken and lymphocytes were extracted, and the expression of Th17 cells in the lymphocytes of different groups of CIA mice was detected by flow Cytometry FCM (FCM). The result is 1: 1. The CIA mouse model was successfully established. Compared with model group, LGIR could improve AI score of CIA mice. Compared with the model group, LGIR could improve the ankle pathological score of CIA mice. Compared with the model group, LGIR could down-regulate the changes of serum IL-23 and IL-17 levels. 5. The ratio of Th17 lymphocyte subsets in the model group was significantly lower than that in the normal control group. The ratio of Th17 cell subsets in the model group and the high dose group was significantly lower than that in the CIA model group. Conclusion the weight LGIR can improve the degree of joint swelling in CIA mice, decrease the levels of IL-23 and IL-17 in serum and reduce the degree of pathological changes of joints. LGIR high dose group can down-regulate the proportion of Th17 cell subsets in lymphocytes of CIA mice.
