应用~1H NMR代谢组技术研究电针治疗胃溃疡的中枢响应机制

【摘要】:目的:采用核磁共振氢谱技术(1H NMR)研究电针胃经穴治疗应激性胃溃疡大鼠大脑皮质、下丘脑、延髓代谢物谱的表达。方法:40只Wistar大鼠随机分为正常组、模型组、胃经组和胆经组。利用束缚冷应激法制备应激性胃溃疡模型大鼠,造模成功后用电针分别刺激胃经组和胆经组相关穴位。收集大鼠大脑皮质、下丘脑、延髓,获取一维1H NMR谱,并利用模式识别方法分析各组间的代谢轮廓差异。结果:模型组与正常组、胃经组、胆经组大鼠大脑皮质、下丘脑和延髓代谢物谱均有明显区分,其中正常组与模型组分离最明显,胃经组最接近正常组,说明其代谢物谱趋向正常。与模型组比较,胃经组大鼠大脑皮质乳酸、萘乙酸、甘露醇等表达上升(P0.05),而肌酸、腺苷、肌苷等表达下降(P0.05),下丘脑乳酸、肌醇、谷氨酸酯等表达上升(P0.05),γ-氨基丁酸等表达下降(P0.05),延髓乳酸、谷氨酸脂、肌醇、丙氨酸等表达上升(P0.05),而萘乙酸、肌酸等表达下降(P0.05)。结论:电针胃经穴可调节胃溃疡大鼠大脑皮质、下丘脑、延髓的代谢产物表达,大脑皮质、下丘脑和延髓是电针胃经穴治疗胃溃疡的中枢响应位点。
[Abstract]:Objective: to study the expression of metabolites in cerebral cortex, hypothalamus and medulla oblongata of rats with stress gastric ulcer treated by electroacupuncture at gastric meridian by 1H NMR. Methods 40 Wistar rats were randomly divided into normal group, model group, gastric meridian group and gallbladder meridian group. The rat model of gastric ulcer induced by stress was prepared by the method of restraint cold stress. After the model was successfully made, the acupoints of gastric meridian group and gallbladder meridian group were stimulated by electroacupuncture respectively. The 1H NMR spectra of cerebral cortex, hypothalamus and medulla oblongata of rats were collected, and the difference of metabolic profile was analyzed by pattern recognition method. Results: the metabolites spectrum of cerebral cortex, hypothalamus and medulla oblongata in model group and normal group, gastric meridian group and gallbladder meridian group were distinguished obviously, among them, the separation between normal group and model group was the most obvious, and the gastric meridian group was closest to normal group. The results showed that the metabolite spectra tended to be normal. Compared with the model group, the expressions of lactic acid, naphthalene acetic acid and mannitol in the gastric meridian group increased, while the expressions of creatine, adenosine and inosine decreased P0.05, lactic acid and inositol in the hypothalamus. The expression of Glutamic acid, 纬 -aminobutyric acid and so on decreased P0.05A, the expressions of lactic acid, glutamate, inositol and alanine increased, while the expressions of naphthyl acetic acid and creatine decreased. Conclusion: electroacupuncture can regulate the expression of metabolites in cerebral cortex, hypothalamus and medulla of rats with gastric ulcer. The central response sites of gastric ulcer treated by electroacupuncture of gastric meridians are cerebral cortex, hypothalamus and medulla oblongata.
