雄激素合成酶在H22荷瘤小鼠不同证候表达差异

【摘要】:目的:研究睾丸雄激素合成酶与肿瘤状态下小鼠证候的关系。方法:采用实验小鼠及其标准化四诊与辨证方法筛选H22荷瘤小鼠邪毒壅盛证、气虚证、阳气虚证、气阴阳虚证,并以Gene Chip Mouse Exon 1.0 ST Array技术检测睾丸组织基因芯片表达谱,重点分析睾丸基因芯片中雄激素合成酶及其调节因子在不同证候小鼠中的差异表达。结果:与正常组比较,Star、Stard5、Cyp17a1、Hsd3b6、Hsd17b3、Sult1e1等基因在早期邪毒壅盛证和气虚证小鼠呈现下调趋势,且在中晚期阳气虚证和气阴阳虚证下调幅度更加显著,Cyp11a1、Hsd3b1、Hsd17b10、Hsd17b11、Hsd17b12、Fdx1、Fdxr、Por、Sult1a1等仅在中晚期阳气虚证和气阴阳虚证中显著下调,Cyp17a1仅在睾丸组织特异性中表达。结论:H22荷瘤小鼠肿瘤形成后,自发产生的虚证越严重,其睾丸雄激素合成酶基因转录活性越低。
[Abstract]:Objective: to study the relationship between testis androgen synthase and syndromes in mice with tumor. Methods: the experimental mice and their four standardized diagnosis and syndrome differentiation methods were used to screen the syndrome of accumulation of evil toxin, deficiency of qi, deficiency of yang and deficiency of qi yin and yang in H22 tumor-bearing mice. Gene Chip Mouse Exon 1.0ST Array technique was used to detect the gene chip expression profile of testis tissue. The differential expression of androgen synthase and its regulatory factors in testicular microarray in mice with different syndromes was analyzed. Results: compared with the normal group, the genes such as Cyp17a1, Hsd3b6, Hsd17b3, Sult1e1 and so on showed a down-regulation trend in the early stage of the mice with the syndrome of excessive accumulation of pathogenic toxin and deficiency of qi. Moreover, the down-regulation of Cyp11a1Hsd3b1C Hsd17b10, Hsd17b11, Hsd17b12, Fdx1FdxrLi, Fdx1FdxrLi, Fdx1a1 and Qi-Yin Yang deficiency syndrome was significantly down-regulated only in testicular tissue specificity in the middle and late stage of Yang Qi deficiency syndrome and Qi Yin and Yang deficiency syndrome, and the down-regulation of Cyp17a1 was only found in testicular tissue. Conclusion the more serious the spontaneous deficiency syndrome is, the lower the transcriptional activity of androgen synthase gene in testis is.
