益气健脾祛痰化瘀法对脾虚痰浊动脉硬化巴马猪水液代谢的影响

【摘要】:目的:观察脾虚痰浊动脉硬化(AS)巴马小型猪水液代谢的变化及益气健脾、祛痰化瘀法的作用。方法:将20头巴马小型猪随机分组,正常组、模型组、益气健脾组及益气健脾祛痰化瘀组各5头。以高脂饮食、跑步劳倦结合冠脉球囊拉伤法制备脾虚痰浊AS病证结合模型至24周。正常组巴马小型猪喂正常饲料,模型组巴马小猪行冠脉球囊拉伤后予高脂饲料结合跑步过劳制备脾虚痰浊AS模型。益气健脾组及益气健脾、祛痰化瘀组在模型组制备基础上,于高脂饲料中添加相应的中药颗粒剂。观察48周后各组巴马小猪的血清钾、钠、氯、尿素氮、肌酐的含量,血清水液代谢调节激素精氨酸加压素(AVP)、心房利钠肽(ANP)、醛固酮(ALD)以及肾组织水通道蛋白(AQP)1、2、3、4、钠钾ATP酶蛋白(Na~+-K~+-ATPase)表达变化。结果:与正常组相比,模型组血清钾、钠、氯、尿素氮、肌酐水平无变化(P0.05),ANP、AVP上调(P0.05),ALD有上调趋势(P0.05),模型组肾组织AQP1、2、3及钠钾ATP酶蛋白表达下调(P0.05),AQP4表达上调。与模型组比较,益气健脾、祛痰化瘀组ANP下调(P0.05),AVP、ALD下调(P0.05),益气健脾组AVP下调(P0.05);益气健脾组及益气健脾、祛痰化瘀组肾组织AQP1、2、3表达均上调(P0.05),AQP4表达下调;益气健脾组钠钾ATP酶蛋白表达上调(P0.05)。结论:脾虚痰浊AS模型未见血清离子紊乱,但水液代谢调节激素中ANP、AVP、ALD有上调,肾组织AQP1、2、3下调,下调,AQP4表达上调;益气健脾及益气健脾祛痰化瘀法可上调肾组织AQP1、2、3,下调AQP4。益气健脾祛痰化瘀法可下调水液代谢调节激素,益气健脾法可上调钠钾ATP酶。益气健脾、祛痰化瘀法在恢复水液代谢调节激素方面优于益气健脾组。
[Abstract]:Objective: to observe the changes of water and liquid metabolism in (AS) small pig with spleen deficiency and phlegm turbid arteriosclerosis and the effect of Qi tonifying spleen removing phlegm and removing blood stasis. Methods: 20 Bama miniature pigs were randomly divided into normal group, model group, invigorating qi group and invigorating spleen group and removing phlegm and removing blood stasis group. The combined model of spleen deficiency and phlegm turbid as disease was established by high fat diet, fatigue of running and balloon injury of coronary artery for 24 weeks. The normal diet was fed to the normal Bama miniature pigs and the model group was fed with high fat diet combined with running overstrain to establish the model of as with spleen deficiency and phlegm turbidity after coronary balloon injury. On the basis of the preparation of the model group, the groups of invigorating qi and invigorating spleen, expelling phlegm and removing blood stasis, added the corresponding traditional Chinese medicine granules in high fat diet. After 48 weeks, serum potassium, sodium, chlorine, urea nitrogen and creatinine were observed in each group of Bama piglets. Serum water solution metabolism regulated the expression of arginine vasopressin (AVP), atrial natriuretic peptide (ANP),) aldosterone (ALD) and renal tissue aquaporin (AQP) (AQP) 1), ATP protein (Na ~ -K ~ -ATPase). Results: compared with normal group, serum potassium, sodium, chlorine, urea nitrogen and creatinine levels in model group showed no change (P0.05). Compared with the model group, ANP was down-regulated (P0.05) and AVP was down-regulated (P0.05) in Qi-invigorating spleen group and dispelling phlegm and removing blood stasis group (P0.05), and the expression of AQP1O2P4 in kidney tissue was down-regulated (P0.05) in invigorating qi group, invigorating qi and invigorating spleen group, dispelling phlegm and removing blood stasis group (P0.05), and down-regulation of AQP4 expression in kidney tissue of Qi-invigorating group and invigorating spleen group (P0.05). The expression of sodium and potassium ATP enzyme protein was up-regulated in invigorating spleen group (P0.05). Conclusion: no serum ion disturbance was found in the model of as with spleen deficiency and phlegm turbidity, but the ALD of ANPP AVPN in water and liquid metabolism regulatory hormone was up-regulated, the expression of AQP4 was down-regulated in renal tissue by AQP1, and the expression of AQP4 was down-regulated by the method of invigorating qi and invigorating spleen and removing phlegm and removing blood stasis in renal tissue. The method of invigorating qi and invigorating spleen and removing phlegm and removing blood stasis can down-regulate the metabolism regulating hormone of water and liquid, and replenish qi and invigorate spleen can upregulate ATP enzyme of sodium and potassium The method of invigorating qi and invigorating spleen and removing phlegm and removing blood stasis is superior to the group of replenishing qi and invigorating spleen.
