壮医药线点灸对佐剂关节炎大鼠滑膜细胞凋亡的影响
[Abstract]:Objective to establish a rat model of rheumatoid arthritis (RA) and to observe the therapeutic effect of moxibustion on synovial cells in rats. Methods the adjuvant arthritis model was established in rats. Methotrexate group (MTX group), Zhuang medicine thread moxibustion group (ZHM group) and combined therapy group (CT group) were given intervention. The index of arthritis and degree of swelling were observed and evaluated; the pathological changes of synovial membrane were observed by HE staining; and the expression of Bcl-2/Bax in synovial tissue was detected by; Western blotting by TUNEL staining. Results the arthritis coefficient in MTX group (P0.05), ZHM group) and CT group (P0.05) was lower than that in model group (RA group). The decrease in CT group was the most significant, and the difference between ZHM group and MTX group was statistically significant (P0.05); HE staining results showed that the proliferation of synovial lining cells and inflammatory cell infiltration in MTX group, ZHM group and CT group were different from RA group). The number of apoptosis in MTX group (P0.05), ZHM group) and CT group (P0.05) was significantly higher than that in RA group. The difference was statistically significant. Compared with MTX group, apoptosis in CT group and ZHM group was significant. The difference was statistically significant (P0.05); MTX group (P0.05), ZHM group, P0.05) and CT group (P0.05) compared with the RA group, the expression of Bcl-2 protein significantly decreased, Bax significantly up-regulated, CT group and ZHM group compared with MTX group Bcl-2 down-regulation, Bax up-regulation was more significant, the difference was statistically significant (P0.05). Conclusion the intervention of Zhuang medicine thread moxibustion can improve the symptoms of adjuvant arthritis rats to a certain extent, and the effect of combined use of methotrexate is more remarkable, and its mechanism may be realized by regulating the apoptosis of synovial cells through Bcl-2 signaling pathway.
