壮医药线点灸对佐剂关节炎大鼠滑膜细胞凋亡的影响

【摘要】：目的通过建立大鼠类风湿关节炎模型,予以壮医药线点灸干预,观察治疗效果,并进一步探讨其对大鼠关节滑膜细胞凋亡的影响。方法建立佐剂关节炎大鼠模型,予以甲氨蝶呤(MTX组)、壮医药线点灸(ZHM组)和两者联合(CT组)干预。观察并评估大鼠关节炎指数和肿胀度;HE染色观察关节滑膜病理变化;TUNEL染色观察大鼠关节滑膜细胞凋亡率;Western blotting检测滑膜组织中Bcl-2/Bax的表达。结果 MTX组(P0.05)、ZHM组和CT组(P0.05)大鼠关节炎系数相比于关节炎模型组(RA组)均有不同程度的降低,其中CT组降低最为显著,ZHM组与MTX组相比差异亦具统计学意义(P0.05);HE染色结果显示MTX组、ZHM组和CT组大鼠滑膜衬里层细胞增生和炎性细胞浸润与RA组相比均有不同程度的改善;MTX组(P0.05)、ZHM组(P0.05)和CT组(P0.05)与RA组相比细胞凋亡数量显著增加,差异具有统计学意义,CT组和ZHM组与MTX组比较,细胞凋亡现象显著,差异具有统计学意义(P0.05);MTX组(P0.05)、ZHM组(P0.05)和CT组(P0.05)与RA组相比大鼠关节滑膜组织中Bcl-2蛋白表达显著下调,Bax显著上调,CT组和ZHM组与MTX组相比Bcl-2下调、Bax上调均更为显著,差异具统计学意义(P0.05)。结论壮医药线点灸干预能够在一定程度上改善佐剂关节炎大鼠症状,联合甲氨蝶呤使用效果更为显著;而且其作用机制可能是通过Bcl-2信号通路调控关节滑膜细胞凋亡来实现的。
[Abstract]:Objective to establish a rat model of rheumatoid arthritis (RA) and to observe the therapeutic effect of moxibustion on synovial cells in rats. Methods the adjuvant arthritis model was established in rats. Methotrexate group (MTX group), Zhuang medicine thread moxibustion group (ZHM group) and combined therapy group (CT group) were given intervention. The index of arthritis and degree of swelling were observed and evaluated; the pathological changes of synovial membrane were observed by HE staining; and the expression of Bcl-2/Bax in synovial tissue was detected by; Western blotting by TUNEL staining. Results the arthritis coefficient in MTX group (P0.05), ZHM group) and CT group (P0.05) was lower than that in model group (RA group). The decrease in CT group was the most significant, and the difference between ZHM group and MTX group was statistically significant (P0.05); HE staining results showed that the proliferation of synovial lining cells and inflammatory cell infiltration in MTX group, ZHM group and CT group were different from RA group). The number of apoptosis in MTX group (P0.05), ZHM group) and CT group (P0.05) was significantly higher than that in RA group. The difference was statistically significant. Compared with MTX group, apoptosis in CT group and ZHM group was significant. The difference was statistically significant (P0.05); MTX group (P0.05), ZHM group, P0.05) and CT group (P0.05) compared with the RA group, the expression of Bcl-2 protein significantly decreased, Bax significantly up-regulated, CT group and ZHM group compared with MTX group Bcl-2 down-regulation, Bax up-regulation was more significant, the difference was statistically significant (P0.05). Conclusion the intervention of Zhuang medicine thread moxibustion can improve the symptoms of adjuvant arthritis rats to a certain extent, and the effect of combined use of methotrexate is more remarkable, and its mechanism may be realized by regulating the apoptosis of synovial cells through Bcl-2 signaling pathway.
