气阴两虚2型糖尿病大鼠氨基酸代谢谱的变化和蛋白干预的影响

【摘要】:目的:研究植物蛋白和动物蛋白对气阴两虚2型糖尿病大鼠氨基酸代谢谱的影响。方法:将110只雄性SD大鼠随机分为空白组(10只),糖尿病模型组(20只),病症结合组(80只),正常组大鼠给予普通饲料,其余给予高糖高脂饲料全程喂养,期间病症结合组灌服青皮附子(15.75 g/kg)每日一次,5w后,腹腔注射小剂量STZ(40 mg/kg)建立实验性糖尿病大鼠模型(正常组除外),将病症结合成模的大鼠按照血糖随机分为气阴两虚糖尿病模型组(附子青皮,15.75 g/kg),大豆蛋白组(大豆蛋白,0.5g/kg),牛奶蛋白组(牛奶蛋白,0.5 g/kg),黄连素组(黄连素,0.1 g/kg),连续灌胃给药4周。蛋白干预4周后,腹主动脉采血分离其血清,1.运用BS-120全自动生化仪检测大生化指标。2.运用1H-NMR寻求对体内特征性代谢物。3.运用AQC柱前衍生高效液相色谱法,利用荧光检测器对血清中游离氨基酸进行分析。结果:蛋白干预4周后,大豆蛋白、牛奶蛋白与黄连素对2型糖尿病大鼠体重和血糖无明显影响。1.与气阴两虚糖尿病组比较,大豆蛋白组总蛋白(TP)、球蛋白(GLB)、血肌酐(Crea)、尿蛋白(Pro)明显升高(P0.05),尿素氮/肌酐(BUN/CREA)、尿素(Urea)显著降低(P0.05),牛奶蛋白组BUN/CREA显著降低(P0.05)。2.与气阴两虚DM模型组比较,大豆蛋白组乳酸、肌酸明显降低(VIP3),α-葡萄糖明显增加且均有显著性差异(VIP3),牛奶蛋白组乳酸明显降低(VIP3),α-葡萄糖明显增加且均有显著性差异(VIP3),黄连素组β-羟异丁酸、乳酸、肌酸明显降低(VIP3),岩藻糖、α-葡萄糖明显增加且均有显著性差异(VIP3)。3与气阴两虚糖尿病模型组比较,大豆蛋白组明显增加丙氨酸、甘氨酸、天冬氨酸、脯氨酸、丝氨酸、谷氨酸、精氨酸、酪氨酸、蛋氨酸、异亮氨酸+亮氨酸、苏氨酸(r0,且VIP1),牛奶蛋白组明显增加丙氨酸、酪氨酸、脯氨酸、苏氨酸、蛋氨酸、苯丙氨酸、赖氨酸、组氨酸、缬氨酸、异亮氨酸+亮氨酸、丝氨酸、甘氨酸(r0,且VIP1),黄连素组明显增加丙氨酸、异亮氨酸+亮氨酸、丝氨酸、牛磺酸、甘氨酸、蛋氨酸、精氨酸、赖氨酸、脯氨酸、组氨酸、苯丙氨酸、缬氨酸、谷氨酸(r0,且VIP1)。结论:蛋白灌胃对气阴两虚2型糖尿病SD大鼠氨基酸代谢能产生了一定的影响,对肝、肾功能具有一定的保护作用。通过代谢组学技术找到标志性差异化合物,从而为2型糖尿病早期预警和气阴虚证侯提供实验依据。
[Abstract]:Aim: to study the effect of plant protein and animal protein on amino acid metabolism in type 2 diabetic rats with deficiency of qi and yin. Methods: 110 male SD rats were randomly divided into control group (n = 10), diabetic model group (n = 20) and disease combination group (n = 80). During the treatment group, aconite was administered once a day (15.75 g/kg), 5 weeks later, An experimental diabetic rat model was established by intraperitoneal injection of small dose STZ (40 mg/kg) (except the normal group). The rats combined with the disease were randomly divided into two groups according to their blood glucose: Qingpi group (qi-yin deficiency, 15.75 g/kg) and soybean protein group (Daqing g/kg). Soybean protein (0.5g/kg), milk protein group (milk protein, 0. 5 g/kg) and berberine group (0. 1 g/kg) were given orally for 4 weeks. After 4 weeks of protein intervention, blood samples from abdominal aorta were collected and their serum was isolated. Using BS-120 automatic biochemical instrument to detect large biochemical indexes. 2. 1H-NMR was used to search for the characteristic metabolites in vivo. The free amino acids in serum were determined by AQC precolumn derivatization HPLC and fluorescence detector. Results: after 4 weeks of protein intervention, soybean protein, milk protein and berberine had no significant effects on body weight and blood glucose of type 2 diabetic rats. 1. Compared with the diabetic group with deficiency of qi and yin, the total protein (TP), globulin (GLB), creatinine (Crea), urinary protein (Pro) in soybean protein group was significantly increased (P0.05), urea nitrogen / creatinine (BUN/CREA), urea (Urea) significantly decreased (P0.05), and BUN/CREA in milk protein group was significantly decreased (P0.05). Compared with the DM model group of deficiency of qi and yin, soybean protein group lactic acid, Creatine (VIP3), 伪 -glucose (VIP3), lactate (VIP3), 伪 -glucose (VIP3), beta-hydroxyisobutyric acid (尾 -hydroxyisobutyric acid), lactic acid (lactic acid) were significantly increased in milk protein group. The levels of creatine (VIP3), alginate, 伪 -glucose and alanine, glycine, aspartic acid, proline, serine, glutamate were significantly increased in the soybean protein group compared with the diabetic model group with deficiency of qi and yin (VIP3), and the levels of alanine, glycine, aspartic acid, proline, serine and glutamate were significantly increased in the soybean protein group. Alanine, tyrosine, proline, threonine, methionine, phenylalanine, lysine, histidine, valine were significantly increased in the arginine, tyrosine, methionine, isoleucine leucine, threonine (r0, and VIP1) groups. Isoleucine leucine, serine, glycine (r0, and VIP1), berberine group significantly increased alanine, isoleucine leucine, serine, taurine, glycine, methionine, arginine, lysine, proline, histidine, phenylalanine, Valine, glutamic acid (r0, and VIP1). Conclusion: protein gavage has a certain effect on amino acid metabolism in type 2 diabetic rats with deficiency of qi and yin, and has protective effect on liver and kidney function. In order to provide experimental basis for early warning of type 2 diabetes mellitus and deficiency of qi and yin syndrome, the marked differential compounds can be found by metabonomics technique.
