回药臭壳虫含药血清对人肺腺癌A549细胞生长增殖的影响

【摘要】:目的:探讨回药臭壳虫(CKC)含药血清对人肺鳞癌A549细胞生长的抑制作用及对细胞形态的影响。方法:将40只雄性SD大鼠按体重随机分为生理盐水对照组(NS)、回药臭壳虫低剂量组(CKCL)、回药臭壳虫中剂量组(CKCM)、回药臭壳虫高剂量组(CKCH)各10只,分别给予相应的CKCL、CKCM、CLCH浓缩水煎剂灌胃,NS组给予等量生理盐水灌胃,连续5天。采用血清药理学方法,制备含药血清;体外培养人肺腺癌A549细胞,将细胞分为实验组与对照组,实验组分别给予10%、20%浓度的相应组大鼠含药血清,对照组给予相同浓度对照组大鼠血清,培养24h、48h、72h后观察细胞形态变化,采用MTT法检测不同浓度含药血清对A549细胞的生长抑制率。结果:光镜下观察发现,各浓度臭壳虫含药血清组细胞形态不规则,胞膜出现不同程度的回缩,胞核固缩,细胞间距增大,其中以20%浓度臭壳虫含药血清高剂量组作用72h最为明显。MTT法检测细胞生长抑制率发现,各实验组不同浓度抑制率均高于对照组(P0.05);回药臭壳虫高剂量组72h抑制率高于低剂量组(P0.05);回药臭壳虫各剂量组72h抑制率高于24h抑制率(P0.05)。结论:回药臭壳虫含药血清可抑制A549细胞的生长增殖。
[Abstract]:Aim: to investigate the inhibitory effect of (CKC) serum on the growth of human lung squamous cell carcinoma cell line A549 and its effect on cell morphology. Methods: forty male SD rats were randomly divided into normal saline control group (n = 10), (NS), control group (n = 10), low dose (CKCL), anther group (n = 10), high dose (CKCM), group (n = 10) and (CKCH) group (n = 10). The rats in the NS group were given the same amount of normal saline for 5 days. Human lung adenocarcinoma cell line A549 was cultured in vitro. The cells were divided into experimental group and control group. The experimental group was given 10% 20% corresponding rat serum. The control group was treated with the same concentration of rat serum and cultured for 24 h or 48 h for 72 h. The growth inhibition rate of A549 cells was detected by MTT assay. Results: under light microscope, it was found that the cell morphology was irregular, the cell membrane was retracted to varying degrees, the nucleus was pyknosis and the cell spacing was increased. Among them, the high dose group with 20% drug serum had the most obvious effect for 72 hours. The inhibition rate of cell growth was detected by MTT assay. The inhibition rate of different concentrations in each experimental group was higher than that of the control group (P0.05); the inhibition rate of 72h in high-dose group was higher than that in low-dose group (P0.05); the inhibition rate of 72h in each dose group was higher than that in 24h (P0.05). Conclusion: the drug serum can inhibit the growth and proliferation of A549 cells.
