电针对功能性消化不良大鼠GAS和SS的影响

【摘要】:目的以功能性消化不良(FD)大鼠为研究对象,观察电针对功能性消化不良模型大鼠一般情况,检测尿D-木糖排泄率、胃蛋白酶含量及胃排空率和小肠推进率,探讨脾虚气滞证候模型大鼠的制备,及电针该模型大鼠胃肠消化功能的影响;检测血清和胃组织中胃泌素(GAS)、生长抑素(SS)的表达,探讨GAS和SS在电针调节胃肠功能中发挥的作用。进一步明确电针治疗FD的作用机理。方法选取24只雄性SD大鼠,按照随机数字表法分为空白组、模型组、电针组,每组8只。除空白组外,其余各组均采用夹尾刺激法配合隔日进食制备FD大鼠模型,造模成功后,电针组给予电针“足三里”及“太冲”治疗,空白组、模型组予束缚固定,每日1次,连续治疗14d。实验过程中观测各组大鼠一般情况和尿D-木糖排泄率,治疗结束后,检测胃排空率,小肠推进率及胃蛋白酶含量,苏木素-伊红(HE)染色观察胃窦组织,双抗体夹心酶联免疫吸附测定法(ELISA)测定血清和胃窦中GAS、SS的表达水平。结果1.一般情况观察比较,造模后,与空白组相比,模型组、电针组可见大鼠烦躁、饮水、饮食减少、毛发枯黄无光泽、大便稀溏,两组鼠体重明显轻于空白组,差异具有统计学意义(p0.05)。电针组经治疗后,一般情况较模型组有所改善,大鼠体重较模型组增长明显,差异具有统计学意义(p0.05),且与空白组差异无统计学意义(p0.05)。2.胃排空率比较,与空白组相比,模型组大鼠的胃排空率降低,差异具显著性差异(p0.01);与模型组相比,电针组大鼠胃排空率增加,差异具有统计学意义(p0.05)。3.小肠推进率比较,与空白组相比,模型组大鼠的小肠推进率显著下降,差异具有统计学意义(p0.01);与模型组相比,电针组大鼠的小肠推进率升高,差异具有统计学意义(p0.05)。4.胃蛋白酶含量比较,与空白组相比,模型组大鼠的胃蛋白酶含量减少,差异具有统计学意义(p0.01);与模型组相比,电针组大鼠的胃蛋白酶含量增加,差异具有统计学意义(p0.05)。5.尿D-木糖排泄率比较,与空白组相比,模型组大鼠的尿d-木糖排泄率降低,差异具有统计学意义(p0.05);与模型组相比,电针组大鼠的尿d-木糖排泄率升高,差异具有统计学意义(p0.05)。6.胃窦和血清中GAS和SS含量比较,与空白组相比,模型组大鼠胃窦和血清中gas含量显著降低,ss含量显著升高,差异均具有统计学意义(p0.01);与模型组相比,电针组大鼠胃窦和血清中gas含量升高,差异具有统计学意义(p0.05),ss含量明显降低,差异具有统计学意义(p0.01)。结论1.通过夹尾刺激+隔日进食的方法造模后,大鼠食量减少、体重下降,胃窦解剖肉眼未见器质性病变,说明造模成功。2.电针组体重、胃排空率、小肠推进率、胃蛋白酶含量、尿D-木糖排泄率都增加,说明电针具有治疗作用,可以改善胃肠道功能。3.电针组胃窦和血清中GAS和SS含量变化,与胃排空率、小肠推进率、胃蛋白酶含量、尿D-木糖排泄率的相关性。电针对GAS、SS表达的调节作用可能是电针治疗FD的机制之一。
