电针对高脂饮食诱导的肥胖小鼠白色脂肪组织中腺苷受体表达的影响
[Abstract]:Aim: to observe the effect of electroacupuncture on adenosine receptor (AR) in white adipose tissue (WAT) of obese (DIO) mice induced by high fat diet, and to explore the mechanism of peripheral action of acupuncture on weight loss. Methods: forty 3-week-old male C 57BL/6 mice were randomly divided into common diet group (n = 12) and hyperlipidemia group (n = 28). DIO model was induced by high-fat diet. Mice were divided into normal control group (n = 5), normal electroacupuncture group (n = 7), model control group (n = 6) and model electroacupuncture group (n = 12). The two electroacupuncture groups were treated 6 times a week for 4 weeks. The body mass of mice was measured and the ratio of lipid to body mass of bilateral epididymal adipose tissue (Epi-WAT) was calculated. The expression of mRNA and protein in Epi-WAT was detected by qPCR,Western blot method. Results: compared with the normal diet group, the body weight of the mice was significantly increased after 12 weeks of high fat feeding (P0.01), in which 18 mice (64.2%) with body mass more than 20% of the normal diet group were compared with the normal control group after the treatment. The body mass of the model control group was still in a higher state (P0.05), the difference of body mass was statistically significant (P0.01), the ratio of Epi-WAT lipid to body mass increased (P0.05); compared with the model control group, The body weight, Epi-WAT lipid / body ratio of the model electroacupuncture group were decreased (P0.05), the body mass change value increased (P0.01). There was no significant difference in A1R mRNA expression in the four groups (P0.05); compared with the normal control group, there was no significant difference in A1R mRNA expression between the four groups (P0.05). The expression of A3R mRNA, A2A R and A2B R protein in the model control group was significantly decreased (P0.01), and the expression of A2A R and A2B) R mRNA and protein in the model electroacupuncture group was higher than that in the model control group (P0.05P 0.01). Conclusion: electroacupuncture can regulate the expression of A2A RGA-A2B R in WAT, which may be the mechanism of electroacupuncture to achieve the effect of weight loss by promoting the metabolism of peripheral WAT.
