背俞指针疗法对GERD大鼠SCF-c-kit-ICC信号通路的影响

【摘要】：目的:通过背俞指针疗法治疗胃食管反流病大鼠,观察SCF-c-kit-ICC信号通路的基因、蛋白表达情况和ICC细胞突起变化,探讨背俞指针疗法对上述变化的影响,以期揭示背俞指针疗法治疗GERD作用机理。方法:将60只GERD大鼠采用随机数字表法,将60只SD大鼠随机平均分为4组,每组15只,空白组和模型组不予干预,参照采用贲门钢圈固定法进行造模,指针治疗组予背俞指针疗法干预治疗,西药对照组予枸橼酸莫沙必利分散片联合兰索拉唑肠溶片灌胃,两组治疗疗程均为2周。干预周期结束后取4组大鼠胃起搏区平滑肌组织若干块,约重100-150mg,活检标本立即投入液氮中,1天后转入-80℃冰箱中保存,另有部分标本置入2.5%戊二醛固定液中保存。本次试验进行检测指标如下:(1)Real-timePCR法观察SCF/c-kit信号通路中SCFmRNA、c-kitmRNA的表达情况;(2)Westernblot法检测GERD模型大鼠胃起搏区平滑肌组织中SCF、c-kit的蛋白质表达情况;(3)透射电镜观察GERD模型大鼠胃起搏区平滑肌组织中Cajal细胞数目及其超微结构的影响。采用SPSS22.0统计软件,通过t检验或X2检验对实验结果进行统计分析。若P0.05时则认为检验结果差异具有统计学意义。结果:1、造模前,各组大鼠体重比较,差异无统计学意义(P0.05)。2、模型组的SCFmRNA和c-kitRNA在胃运动起搏区平滑肌组织的表达较空白组下调(P0.05),具有统计学意义;指针治疗组和西药对照组的SCFmRNA和c-kitmRNA在胃运动起搏区平滑肌组织的表达较模型组显著升高(P0.01),具有统计学意义;指针治疗组和西药对照组的SCFmRNA和c-kitmRNA在胃运动起搏区平滑肌组织的表达相比无明显差异(P0.05),无统计学意义,并与正常组相比无显著性差异(P0.05),无统计学意义。3、GERD大鼠模型组的SCF和c-kit在胃运动起搏区平滑肌组织的蛋白表达较空白组下调(P0.05),具有统计学意义;指针治疗组和西药对照组的SCF和c-kit在胃运动起搏区平滑肌组织的蛋白表达较模型组显著升高(P0.01),具有统计学意义;指针治疗组和西药对照组的SCF和c-kit在胃运动起搏区平滑肌组织的蛋白表达相比无明显差异(P0.05),无统计学意义,并与正常组相比无显著性差异(P0.05),无统计学意义。4、模型组ICC细胞数量和超微结构明显改变,突起数量明显减少,突起长度减短,与平滑肌细胞之间的缝隙连接明显减少,且线粒体大多呈空泡样改变。指针治疗组、西药对照组和模型组相比,ICC细胞数量和超微结构有明显改善,以及与平滑肌细胞之间的细胞突起数量及长度、缝隙连接有明显改善。结论:背俞指针疗法可能通过调控SCF-c-kit-ICC信号通路的mRNA基因水平的表达、蛋白水平的表达及细胞形态的改变,促进GERD的胃肠动力恢复。

The purpose of this study was to reveal the mechanism of the treatment of GERD with back-Shu pointer therapy. Methods: sixty GERD rats were randomly divided into 4 groups (15 rats in each group) by random digital table method. The blank group and model group were not interfered, and the model was made with the method of cardia ring fixation. The control group was treated with mosapride citrate dispersible tablets combined with lansoprazole enteric-coated tablets for 2 weeks. At the end of the intervention cycle, a number of smooth muscle tissues of gastric pacing area were taken from four groups of rats, weighing about 100-150 mg. The biopsy specimens were immediately injected into liquid nitrogen, and then transferred to -80 鈩，
