温阳益气方对溃疡性结肠炎大鼠Treg细胞的免疫调节作用

【摘要】:目的:观察温阳益气方对由三硝基苯磺酸(TNBS)/乙醇诱导的溃疡性结肠炎(UC)大鼠模型Treg细胞的影响,进而探讨温阳益气方治疗UC的作用机制和靶点。方法:经过TNBS/乙醇造模成功后,分为空白组、温阳益气(WYYQ)组、模型组、美沙拉嗪(MT)组。用灌胃的方式给药持续14天后,把大鼠处死并取标本。观察UC大鼠结肠粘膜组织的病理改变采用苏木精-伊红染色(HE)法。检测大鼠血清中IL-10的表达用酶联免疫吸附试验(ELISA);检测特异性转录因子Foxp3mRNA的基因水平用实时荧光定量PCR法。结果:1.与空白组相比,模型组大鼠出现神态萎靡、反应迟钝、弓背蜷缩、毛质稀松、活动极少、体温降低、便溏、进食不佳、体重减轻等一系列类似阳虚的表现。2.HE染色后电镜下病理切片结果示:与空白组相比,模型组大鼠的结肠粘膜水肿、充血、糜烂明显,腺体排列紊乱甚至断裂消失,炎细胞(中性粒细胞、淋巴细胞)浸润严重,粘膜上皮组织严重损伤,多处隐窝溃疡形成。病理组织评分模型组与空白组相比分值显著增高,且差异有统计学意义(P0.05);MT组和WYYQ组的分值与模型组相比显著降低,且差异有统计学意义(P0.05)。3.与空白组相比,大鼠血清中IL-10含量在模型组中浓度显著降低,且差异有统计学意义(P0.05);MT组和WYYQ组与模型组相比较,大鼠血清中IL-10的浓度含量显著增高,且差异有统计学意义(P0.05);MT组大鼠血清中IL-10的浓度含量较WYYQ组低,且差异有统计学意义(P0.05)。4.与空白组相比,模型组中Foxp3mRNA在大鼠结肠组织中的含量表达显著减少,且差异有统计学意义(P0.05);MT组和WYYQ组结肠组织中Foxp3 mRNA含量表达与模型组相比,水平显著增高,且差异有统计学意义(P0.05)结论:温阳益气方可通过增加血清中IL-10的含量,上调结肠组织中Foxp3的表达,增加机体Treg细胞含量,提高机体免疫力,从而有利于治疗UC。
[Abstract]:Aim: to observe the effect of Wenyang Yiqi recipe on Treg cells of (UC) rat model of ulcerative colitis induced by trinitrobenzene sulfonic acid (TNBS) / ethanol, and to explore the mechanism and target of WYYQF in the treatment of UC. Methods: after TNBS/ ethanol was successfully established, the rats were divided into three groups: blank group, (WYYQ) group, model group and (MT) group. Rats were killed and specimens were taken after 14 days of gavage. The pathological changes of colonic mucosa in UC rats were observed by hematoxylin-eosin staining (HE) method. The expression of IL-10 in rat serum was assayed by enzyme linked immunosorbent assay (ELISA);) to detect the gene level of specific transcription factor (Foxp3mRNA) by real-time fluorescence quantitative PCR method. Results: 1. Compared with the blank group, the rats in the model group appeared to be sluggish, slow to react, curled up on the back of the bow, fluffy hair, very little activity, hypothermia, loose stools, and poor food intake. 2.HE staining showed that compared with the blank group, the colonic mucosal edema, hyperemia, erosion, gland arrangement disorder and even rupture disappeared in the model group. Inflammatory cells (neutrophils, lymphocytes) infiltration, mucosal epithelial tissue damage, many recess ulcer formation. The pathological tissue score in the model group was significantly higher than that in the blank group, and the difference was statistically significant (P0.05). (P0.05) the score of); MT and WYYQ group was significantly lower than that of the model group, and the difference was statistically significant (P0.05). Compared with the blank group, the concentration of serum IL-10 in the model group decreased significantly, and the difference was statistically significant (P0.05). Compared with model group, the concentration of IL-10 in serum of MT group and WYYQ group was significantly higher than that of model group (P0.05). The concentration of IL-10 in serum of MT group was lower than that of WYYQ group, and the difference was statistically significant (P0.05). Compared with the blank group, the expression of Foxp3mRNA in the colonic tissue of the model group was significantly decreased, and the difference was statistically significant (P0.05). The expression of Foxp3 mRNA in colon tissue of MT group and WYYQ group was significantly higher than that of model group, and the difference was statistically significant (P0.05) conclusion: Wenyang Yiqi decoction could increase the content of IL-10 in serum. The expression of Foxp3 in colon tissue is up-regulated, the content of Treg cells is increased, and the immunity of the body is enhanced, which is beneficial to the treatment of UC..
