电针抗高脂饮食诱导肥胖小鼠的作用及对脂肪组织炎症相关因子表达的影响

【摘要】:目的:观察电针抗高脂饮食诱导肥胖小鼠的作用及对脂肪组织炎症相关因子表达的影响。方法:选用40只刚断乳的SPF级C57BL/6J雄性小鼠,随机挑选8只普通饲料喂养为正常组,另一组32只高脂饲料喂养,将造模成功的16只随机分为模型组和电针组。比较各组小鼠体重、体脂、血脂等相关指标的差异,用HE染色观察比较各组小鼠脂肪的形态学差异,用实时荧光定量PCR检测各组小鼠脂肪组织中IL-6、TNF-α与MCP-1表达变化。结果:与模型组相比,电针组小鼠体重、内脏脂肪量下降明显;胆固醇、低密度脂蛋白含量均显著下降;脂肪组织IL-6、TNF-α、MCP-1的mRNA表达水平亦显著下降。结论:电针可以降低肥胖小鼠的体重、体脂,血脂,抑制肥胖小鼠脂肪组织炎症因子IL-6、TNF-α、MCP-1 mRNA的表达,改善肥胖小鼠炎症反应状态,从而实现减肥疗效。
[Abstract]:Aim: to observe the effect of electroacupuncture on obesity induced by high fat diet and the expression of inflammatory factors in adipose tissue. Methods: 40 newly weaned SPF grade C57BL/6J male mice were randomly selected as normal diet group and 32 high fat diet groups. The 16 mice were randomly divided into model group and electroacupuncture group. The relative indexes such as body weight, body fat and blood lipid in each group were compared. The morphologic difference of fat in each group was observed by HE staining, and the IL-6, in adipose tissue of each group was detected by real-time fluorescence quantitative PCR. The expression of TNF- 伪 and MCP-1 was changed. Results: compared with the model group, the body weight, visceral fat content, cholesterol and low density lipoprotein contents in the electroacupuncture group were significantly decreased, and the mRNA expression levels of IL-6,TNF- 伪 and MCP-1 in adipose tissue were also significantly decreased. Conclusion: electroacupuncture can reduce the body weight, body fat, blood lipid, inhibit the expression of IL-6,TNF- 伪 and MCP-1 mRNA in adipose tissue of obese mice, improve the inflammatory reaction state of obese mice, and achieve the effect of weight loss.
