槲皮素脂质体对糖尿病大鼠肾脏糖基化终产物及其受体和TGF-β1表达的影响

【摘要】:目的:观察槲皮素脂质体(LQ)对糖尿病大鼠肾脏糖基化终产物(AGEs)及其受体(RAGE)和TGF-β1表达的影响。方法:采用旋转蒸发法制备槲皮素脂质体,高糖高脂饲料联合腹腔注射链脲佐菌素(STZ)建立2型糖尿病大鼠模型,并随机分为糖尿病模型组(DM组),槲皮素脂质体低(LQ-L)、中(LQ-M)、高(LQ-H)剂量组,氨基胍(AG)对照组(AG组),另设正常组(N组)。灌胃治疗8周后测定各组大鼠血糖、体质量、肾脏肥大指数(KI)、血尿素氮(BUN)、血肌酐(Scr)、血清中超氧化物歧化酶(SOD)、丙二醛(MDA),ELISA法测血清AGEs表达和24 h尿微量白蛋白,PAS染色观察肾脏病理改变,免疫组化测肾组织AGEs和TGF-β1表达,RT-PCR检测肾皮质RAGE mRNA和TGF-β1 mRNA表达水平。结果:与N组比较,DM组大鼠血糖、KI、BUN、Scr、MDA含量、血清AGEs和24 h尿微量白蛋白显著升高,血清中SOD活性水平、体质量明显降低;肾小球体积萎缩,基底膜增厚;肾组织AGEs、TGF-β1和RAGE mRNA、TGF-β1 mRNA表达增高。与DM组比较,LQ各剂量组大鼠血糖、KI、BUN、Scr、MDA含量、血清AGEs和24 h尿微量白蛋白均降低,血清中SOD活性水平、体质量增加;病理改变明显减轻;肾组织AGEs、TGF-β1和RAGE m RNA、TGF-β1 mRNA表达降低,以中剂量组作用更明显。结论:LQ可抑制蛋白质非酶糖基化反应和细胞外基质(ECM)增生,从而减少肾组织AGEs、TGF-β1生成及RAGE mRNA、TGF-β1 mRNA表达水平,对糖尿病大鼠肾脏具有保护作用。
[Abstract]:Aim: to observe the effect of quercetin liposome (LQ) on the expression of (AGEs), its receptors (RAGE) and TGF- 尾 1 in diabetic rats. Methods: quercetin liposomes were prepared by rotating evaporation. The rats with type 2 diabetes were induced by high glucose and high fat diet and intraperitoneal injection of streptozotocin (STZ). The rats were randomly divided into diabetic model group (DM group). Quercetin liposome (LQ-L), moderate (LQ-M), high dose (LQ-H) group, aminoguanidine (AG) control group (AG group), and normal group (N group). Blood glucose, body mass, renal hypertrophy index (KI), blood urea nitrogen (BUN), serum creatinine (Scr), superoxide dismutase (SOD),) and malondialdehyde (SOD),) (MDA), in rats were measured after 8 weeks of gastric perfusion. The expression of serum AGEs and 24 h urinary microalbumin were measured by ELISA method, the pathological changes of kidney were observed by PAS staining, the expressions of AGEs and TGF- 尾 1 in renal tissues were detected by immunohistochemistry, and the expressions of RAGE mRNA and TGF- 尾 1 mRNA in renal cortex were detected by RT-PCR. Results: compared with group N, the contents of blood glucose, KI,BUN,Scr,MDA, serum AGEs and 24 h urinary microalbumin in DM group were significantly increased, the activity of SOD in serum and body weight were significantly decreased, glomerular volume atrophy and basement membrane thickening were observed. The expression of AGEs,TGF- 尾 1 and RAGE mRNA,TGF- 尾 1 mRNA was increased in renal tissue. Compared with DM group, the levels of blood glucose, KI,BUN,Scr,MDA, serum AGEs and 24 h urinary microalbumin decreased, the activity of SOD in serum and body mass increased, and the pathological changes were alleviated. The expression of AGEs,TGF- 尾 1 and RAGE m RNA,TGF- 尾 1 mRNA in renal tissue was decreased, especially in middle dose group. Conclusion: LQ can inhibit the protein non-enzymatic glycosylation reaction and the proliferation of extracellular matrix (ECM), thus reduce the production of AGEs,TGF- 尾 1 and the expression of RAGE mRNA,TGF- 尾 1 mRNA in renal tissue. It has protective effect on the kidney of diabetic rats.
