高脂诱导自发性高血压大鼠脂肪组织SIRT1的表达下调及清肝滋肾法的调控作用

【摘要】:目的:探讨清肝滋肾中药(针箭颗粒Ⅱ号)通过上调内脏脂肪组织中SIRT1的表达改善高血压代谢紊乱的作用及机制。方法:通过高脂饮食诱导获得自发性高血压大鼠代谢紊乱模型。选取17只4周龄的雄性自发性高血压大鼠(SHR),分为高脂饮食组12只和正常饮食组5只,正常饮食组给予基础饲料,高脂饮食组给予高脂饲料。高脂饮食组则随机分为针箭颗粒组6只(22.5g/kg/d,灌胃)和模型对照组6只(同体积蒸馏水,灌胃)。每周测量体重、血压2次,根据体重改变药物给药量。给药9周后处死,获取大鼠内脏脂肪(附睾周围)并称重,部分内脏脂肪分离脂肪细胞,部分提取mRNA和蛋白,部分给予4%多聚甲醛固定HE染色。1)通过定期监测SHR鼠血压、体重,评估SHR大鼠代谢情况。2)采用RT-PCR和Western Blot法测定内脏脂肪组织SIRT1 mRNA-及蛋白、裂解酶Caspase-1蛋白的表达水平,评估SIRT1表达水平和活性的变化;结果:1.清肝滋肾中药(针箭颗粒Ⅱ号)对SHR大鼠体重的影响:基线时SHR大鼠三组之间体重无明显差异(P0.05);干预9周后模型组体重显著高于对照组(P0.01),针箭颗粒组体重显著低于模型组(P0.01),对照组与针箭颗粒组之间无显著差异(P0.05)。2.清肝滋肾中药(针箭颗粒Ⅱ号)对SHR大鼠血压的影响:治疗前模型组SBP高于对照组(P0.05),其余组间无显著差异(P0.05);三组间DBP无明显差异(P0.05)。干预9周后,模型组与对照组之间血压(SBP、DBP)无明显差异,针箭颗粒组虽有下降趋势但无统计学差异(P0.05)。3.清肝滋肾中药(针箭颗粒Ⅱ号)对SHR大鼠内脏脂肪(附睾周围)的影响3.1各组附睾周围脂肪重量及其体脂比的比较:1)干预9周后模型组附睾周围脂肪含量高于对照组(P0.05),但针箭颗粒组与模型组间无明显差异(P0.05);2)干预9周后模型组体脂比显著高于对照组(P0.01),但针箭颗粒组与模型组间无明显差异(P0.05)。3.2镜下评估附睾周围脂肪体积及脂滴含量:模型组与对照组相比,脂肪细胞体积增大,脂滴含量增加;针箭颗粒组与模型组相比,脂肪细胞体积减小,脂滴含量减少。4.清肝滋肾中药(针箭颗粒Ⅱ号)对SHR大鼠内脏脂肪(附睾周围)组织SIRT1表达及活性变化的影响4.1清肝滋肾中药(针箭颗粒Ⅱ号)对附睾周围脂肪组织中SIRT1 mRNA表达的影响:模型组SIRT1 mRNA的表达显著低于对照组(P0.01),而针箭颗粒组SIRT1 mRNA的表达高于模型组(P0.05)。4.2清肝滋肾中药(针箭颗粒Ⅱ号)对附睾周围脂肪组织中SIRT1蛋白表达的影响:干预9周后,模型组SIRT1蛋白表达显著低于对照组(P0.01),针箭颗粒组SIRT1蛋白表达则高于模型组(P0.05)。4.3清肝滋肾中药(针箭颗粒Ⅱ号)对附睾周围脂肪组织中裂解酶Caspase1蛋白表达的影响:干预9周后三组之间Caspase1蛋白表达水平无显著差异(P0.05)。结论:1.清肝滋肾中药(针箭颗粒Ⅱ号)可降低SHR合并代谢紊乱大鼠体重,减少内脏脂肪的堆积。2.清肝滋肾中药(针箭颗粒Ⅱ号)可能是通过上调内脏脂肪组织中SIRT1的表达来改善高血压代谢紊乱。
[Abstract]:Objective: To study the effect and mechanism of the expression of SIRT1 in visceral fat tissue by regulating the expression of SIRT1 in visceral fat tissue. Methods: The metabolic disorder model of spontaneously hypertensive rats was induced by high-fat diet. 17-week-old male spontaneously hypertensive rats (SHR) were selected and divided into two groups: high-fat diet group (n = 12) and normal diet group (n = 5). The normal diet group was given the basic feed and the high-fat diet group was given a high-fat diet. The high-fat diet group was randomly divided into 6 groups (22.5 g/ kg/ day, gavage) and 6 model control groups (with the same volume of distilled water and intragastric administration). Body weight was measured weekly, blood pressure was 2 times, and the amount of drug was changed according to body weight.
