针刺对糖尿病抑郁大鼠血糖、情绪昼夜节律及Per1mRNA、Per2mRNA表达的影响

【摘要】:目的:本实验应用行为学观察、时间生物学研究方法及分子生物学技术等,研究糖尿病抑郁大鼠的血糖-HPA轴-情绪昼夜节律的改变特点,并且探讨针刺对Per1mRNA、 Per2mRNA的基因表达的影响,揭示针刺调节糖尿病抑郁的血糖-HPA轴-情绪昼夜节律紊乱的相关分子生物机理。方法:将32只雄性SD大鼠,随机分为空白组、糖尿病组、糖尿病抑郁组及针刺组,每组8只。空白组给予普通喂养;糖尿病组给予高脂喂养及STZ试剂腹腔注射造模;糖尿病抑郁组给予高脂喂养、STZ试剂腹腔注射及慢性应激造糖尿病抑郁模型;针刺组在糖尿病抑郁组造模基础上针刺后三里、三阴交及百会穴,留针15分钟,连续治疗7天。针刺治疗后,采用股动脉取血及断颈取脑法取出各组大鼠的血清及下丘脑组织,采用酶联免疫吸附法测定皮质醇水平,采用实时定量PCR技术对大鼠SCN内的的per2mRNA基因的表达量进行检测。结果:1.糖尿病组血糖及情绪: (1)与空白组比较,糖尿病组大鼠的血糖值升高(P0.05),旷场实验数据值下降(P0.05)。(2)与空白组比较,糖尿病组大鼠的24小时内的血糖谷峰值差异大(P0.05);且其24小时内的血糖值及旷场实验数据的峰值出现时间延后。2.糖尿病抑郁组血糖及情绪: (1)与空白组大鼠及糖尿病组大鼠比较,糖尿病抑郁组大鼠的血糖值升高(P0.05),旷场实验数据值下降(P0.05)。 (2)与空白组比较,糖尿病抑郁组大鼠24小时内血糖的谷峰值差异大(P0.05),糖尿病抑郁组大鼠24小时内的血糖峰值出现时间延后,其24小时内的旷场实验数据值改变趋势与空白组大鼠相反。3.针刺组血糖及情绪: (1)与糖尿病抑郁组比较,针刺组大鼠的血糖值下降(P0.05),其旷场实验数据值上升(P0.05)。 (2)针刺组大鼠的血糖谷峰值差异大(P0.05),针刺组大鼠血糖值的次高峰与次低谷的出现时间与空白组相同,其峰谷值出现时间有向空白组移位的趋势,且10:00-22:00的最大值与最小值无统计学意义(P0.05);针刺组大鼠旷场实验数据与空白组均呈现双峰双谷的节律变化。4.各组的皮质醇水平:与空白组比较,糖尿病组大鼠、糖尿病抑郁组大鼠及针刺组大鼠的皮质醇水平升高(P0.05);且三组的皮质醇比较:糖尿病抑郁组糖尿病组针刺组(P0.05)。5.各组的Per1mRNA、Per2mRAN基因表达:与空白组比较,糖尿病组大鼠、糖尿病抑郁组大鼠及针刺组大鼠的Per2mRNA的表达量升高(P0.05);与糖尿病抑郁组及糖尿病组大鼠比较,针刺组大鼠的Per2mRNA的表达量下降;各组大鼠的Per1mRNA的表达量比较:空白组针刺组糖尿病组糖尿病抑郁组P0.05)。结论:1、针刺具有改善糖尿病抑郁大鼠的血糖、抑郁情绪及调节其血糖、情绪节律的作用。2、针刺具有降低糖尿病抑郁大鼠的皮质醇水平的作用。3、针刺具有调节糖尿病抑郁大鼠的Per2mRNA的表达的作用;针刺可能是通过调节Per2mRNA基因的表达,从而调节血糖及情绪节律紊乱,以治疗糖尿病抑郁的。
[Abstract]:Objective: to study the changes of blood glucose-HPA axis-mood circadian rhythm in diabetic depression rats by behavioral observation, time biology and molecular biology, and to explore the effect of acupuncture on Per1mRNA,. The effect of Per2mRNA gene expression on the molecular biological mechanism of acupuncture regulating blood glucose-HPA axis-emotional circadian rhythm disorder in diabetic depression was revealed. Methods: 32 male SD rats were randomly divided into blank group, diabetic depression group and acupuncture group with 8 rats in each group. The diabetic depression model was established by high fat feeding and intraabdominal injection of STZ reagent, high fat feeding, intraabdominal injection of STZ reagent and chronic stress in diabetic depression group. On the basis of modeling in diabetic depression group, acupuncture group was treated with Sanli, Sanyinjiao and Baihui points for 15 minutes for 7 days. After acupuncture treatment, the serum and inferior colliculus brain tissue of rats in each group were taken out by the method of taking blood from femoral artery and severing neck, and the level of cortisol was measured by enzyme-linked immunosorbent assay (Elisa). The expression of per2mRNA gene in rat SCN was detected by real-time quantitative PCR. Results: 1. Blood glucose and emotion in diabetic group: (1) compared with the blank group, the blood glucose value in the diabetic group was higher than that in the blank group (P 0.05), and the data value in the open field experiment (P 0.05). (2) was lower than that in the blank group. The peak value of blood glucose in diabetic group was significantly different within 24 hours (P 0.05). The peak value of blood glucose and open field experimental data within 24 hours was delayed. 2. Blood glucose and emotion in diabetic depression group: (1) compared with blank group and diabetic group, the blood glucose value of diabetic depression group was higher than that of blank group and diabetic group (P 0.05), and the data value of open field experiment was decreased (P 0.05). (2) compared with the blank group, the peak value of blood glucose in the diabetic depression group was significantly different from that in the blank group within 24 hours (P 0.05), and the peak value of blood glucose in the diabetic depression group was delayed within 24 hours. The change trend of open field experimental data within 24 hours was opposite to that of blank group. 3. Blood glucose and emotion in acupuncture group: (1) compared with diabetic depression group, the blood glucose value of acupuncture group decreased (P 0.05), and the data value of open field experiment increased (P 0.05). (2) the peak value of blood glucose in acupuncture group was significantly different (P 0.05). The secondary peak and subtrough of blood glucose value in acupuncture group were the same as those in blank group, and the appearance time of peak and valley value tended to shift to blank group. There was no significant difference between the maximum value and the minimum value of 10 鈮,
