黎药益智仁不同极性部位提取物对实验性肠炎模型小鼠的影响

【摘要】:目的:考察黎药益智仁4种不同极性部位提取物对实验性肠炎模型小鼠的影响。方法:将益智仁70%乙醇提取物浸膏用水分散后,依次用石油醚、三氯甲烷、乙酸乙酯和正丁醇提取得相应部位提取物。将42只小鼠随机分为造模组(36只)和空白组(6只),造模组大鼠采用2,4,6-三硝基苯磺酸法复制实验性肠炎模型。成模后,将造模小鼠随机分为模型组、阳性组(柳氮磺胺吡啶肠溶片,52 g/kg)和益智仁不同极性部位(石油醚、三氯甲烷、乙酸乙酯和正丁醇部位)提取物组(剂量均为10 g/kg,以生药计),每组6只,ig给药,每天1次,连续9 d,给药体积为0.2 m L/10 g;空白组和模型组小鼠ig生理盐水。测定小鼠体质量变化,对疾病活动指数(DAI)进行评分,测定结肠组织中髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)、丙二醛(MDA)活性,观察结肠组织病理变化并进行评分。结果:与空白组比较,模型组小鼠体质量下降百分率、DAI评分以及结肠组织中MPO、MDA活性均显著升高(P0.05或P0.01),结肠组织发生明显病变。与模型组比较,各给药组小鼠体质量显著回升,DAI评分显著降低,MPO活性显著降低,病理评分显著降低;阳性组、益智仁正丁醇提取物和乙酸乙酯提取物组小鼠结肠组织中SOD活性显著升高,阳性组和益智仁乙酸乙酯提取物组小鼠结肠组织中MDA活性显著降低,差异均有统计学意义(P0.05或P0.01)。结论:益智仁4种极性部位提取物对实验性肠炎模型小鼠均有一定改善作用,其中以乙酸乙酯部位和正丁醇部位效果较好。

Methods: the 70% ethanol extract of Yizhiren was dispersed with water and then extracted with petroleum ether, trichloromethane, ethyl acetate and n-butanol.
