益气解毒方对肾癌干细胞增殖及凋亡影响的实验研究

【摘要】：目的:明确益气解毒方对ACHN肾癌干细胞的增殖及凋亡的影响。方法:采用高、中、低3种浓度的“益气解毒方”药物血清干预体外培养的ACHN肾癌干细胞。1.采用二甲基偶氮唑蓝(MTT)实验检测“益气解毒方”药物血清对ACHN肾癌干细胞增殖的影响;2.流式细胞仪检测“益气解毒方”药物血清干预下ACHN肾癌干细胞的凋亡;3.流式细胞仪检测“益气解毒方”药物血清干预下ACHN肾癌干细胞P53蛋白的表达情况。结果:1.“益气解毒方”药物血清可以抑制ACHN肾癌干细胞的增殖,各组抑制率分别为:空白对照组24 h:0.36%,48h:0.41%,72:0.43%;低浓度组24 h:15.7%,48h:32.7%,72h:44.3%;中浓度组24h:36.7%,48h:53.3%,72h:63.7%;高浓度组24h:53.3%,48h:67%,72h:77.7%;各组间比较P值均0.05,差异有统计学意义。2.三种浓度“益气解毒方”药物血清下均可诱导ACHN肾癌干细胞的凋亡,各组凋亡率分别为:空白对照组0.55±0.075%;低浓度组:3.48±0.505%;中浓度组:9.33±0.612%;高浓度组:18.36±1.511%,各组间比较P值均0.05,差异有统计学意义。3.三种浓度的“益气解毒方”药物血清均不同程度的上调了ACHN肾癌干细胞的P53蛋白的表达,分别为空白对照组:0.78±0.25%;低浓度组:7.24±0.80%;中浓度组:11.72±1.19%;高浓度组:17.68±1.06%;各组间比较P值均0.05,差异有统计学意义。结论:1.“益气解毒方”药物血清可以抑制ACHN肾癌干细胞增殖。2.“益气解毒方”药物血清可以诱导ACHN肾癌干细胞凋亡。3.“益气解毒方”药物血清可以提高ACHN肾癌干细胞P53蛋白的表达。
[Abstract]:Objective: to investigate the effect of Yiqi jiedu recipe on proliferation and apoptosis of ACHN renal cell carcinoma stem cells. Methods: high, medium and low concentrations of "Yiqi jiedu recipe" were used to intervene ACHN renal cell carcinoma stem cells in vitro. The effect of "Yiqi jiedu recipe" on the proliferation of ACHN renal cell carcinoma stem cells was detected by dimethyl azo blue (MTT) assay. Apoptosis of ACHN renal cell carcinoma stem cells was detected by flow cytometry under the intervention of "Yiqi jiedu recipe" serum.
